摘要
目的 :应用人结肠癌建株LoVo细胞接种裸鼠 ,鼠间移植传代 ,建立人结肠癌裸鼠移植瘤模型。方法 :观察移植瘤大体形态和组织病理学检查 ,测定其肿瘤组织与宿主血液中的癌胚抗原 (CEA)值 ,并应用免疫组化观察肿瘤细胞中CEA分布以及153 Sm标记抗CEA单抗的荷瘤裸鼠体内放射免疫显像。结果 :移植瘤的形态和功能特性与原发肿瘤基本相似 ,移植瘤的移植成功率为 10 0 % ;放免显像显示标记抗体能浓聚于肿瘤部位 ,经腹腔注射标记抗体后 48h ,肿瘤显像清晰 ,肿瘤组织 /血液放射性比值 (T/B)为 1.42。结论
?Objective:To establish human colonic carcinoma xenograft model in nude mice with human colonic carcinoma LoVo cells inoculation and subgraft through mice.Methods:The morphological and histopathological features of transplanted tumor were examined.Specimens of tumor tissue and blood samples taken from the hosts were assayed for CEA levels respectively.Immunohistochemical localization of CEA in the tumor tissues was also determined.Radioimmunoimaging studies of the nude mice bearing transplanted tumors with 153 Sm labeled CEA McAb were taken.Results:It showed that the morphological and functional characteristics of the transplanted tumors were strikingly identical to the original human tumor.The successful transplantation rate was 100%.After injection of 153 Sm CEA McAb intraperitoneally to nude mice xenografted with human colonic carcinoma,labeled antibody was found to be concentrated preferentially in the tumor.The ratio of radioactivity in tumor and blood was 1.42 after 48 hours following administration.Conclusions:The nude mice model of human colonic carcinoma so established provides an ideal tumor model for in vivo radioimmunoimaging and radioimmunotherapy. 〔
