摘要
探索了影响樱花细胞悬浮培养的因素,优化了其液体培养基不同激素配比、培养物密度、培养时间、摇床转速等培养条件,以建立樱花细胞悬浮培养体系。结果表明:细胞悬浮培养的最佳条件是MS+1.0mg/L6-BA+2.0mg/LNAA+1.0mg/L2、4-D液体培养基中,于26℃、120r/min的摇床上,细胞悬浮培养细胞最佳起始密度为2.5×104个·mL-1时,培养14d,可得到稳定的悬浮细胞系。
This experiment explores the influencing factors of Prunus campanulata cells suspension culture, optimizes the culture conditions, such as different hormone proportion of liquid medium, the training content density and training time,etc. ,in order to set up the cell suspension culture system of cherry blossom. The results show that :when the cells are cultured in 4--D liquid medium MS+1.0mg/L6-BA+2.0mg/LNAA+1.0mg/L2、4-D,at 26℃on the 120 r/min wave bed,and the cells optimum starting density of cell suspension culture is 2.5 ×10^4 o mL^-1,and lasts 14 days,it can get stable suspension cell lines,therefore,it can be considered as the best condition of cell suspension culture.
