摘要
目的:探讨促性腺激素和细胞因子在上皮性卵巢癌发生发展中的作用。方法:应用四甲基偶氮唑蓝(MTT)实验观察卵泡刺激素(FSH)、黄体生成素(LH),人重组白介素-6(hrIL–6)对卵巢癌细胞株增殖的影响。结果:卵泡刺激素、黄体生成素及人重组白介素-6对卵巢癌细胞株人卵巢低分化乳头状腺癌细胞株(OVCAR-3)、人上皮性卵巢癌细胞株(Caov-3)具有促增殖作用并呈剂量依赖性,与对照组相比差异有显著意义。在卵巢癌细胞培养液中加入50μg/ml抗白介素-6(IL-6)中和性抗体,两种卵巢癌细胞的生长均有所减慢。结论:肌体内促性腺激素水平异常与上皮性卵巢肿瘤形成密切相关,激素与细胞因子影响其发展和转归。
Objective: To investigate the role of gonadotropins and cytokines in the development of epithelial ovarian carcinoma.Methods: Two epithelial ovarian carcinoma cell lines(OVCAR-3,Caov-3) were cultured with follicle-stimulating hormone(FSH),luteinizing hormone(LH) and human recombinant interleukin-6(hrIL-6),respectively.The degree of proliferation of two cell lines was determined by MTT.Results:The proliferation was increased by gonadotropins or IL-6 for a period of 96 h(P〈0.05 vs control group) and a dose-dependent effect was seen in both OVCAR-3 and Caov-3.Most significantly,for both cell lines,50 μg/ml anti-IL-6 antibody partially inhibited the hormone-stimulated cell proliferation.Conclusion: Elevated serum gonadotropin is one of the factors contributing significantly to the development of epithelial ovarian carcinoma.Gonadotropins and cytokines are sensitive biomarkers for for the prognosis of the ovarian carcinoma.
