地塞米松、齐留通对哮喘大鼠5-脂氧合酶的调节 被引量:2

摘要 目的探讨哮喘大鼠中地塞米松、齐留通与5-脂氧合酶(5-LOX)基因、蛋白表达的关系。方法制备卵蛋白致敏哮喘大鼠模型;通过原位杂交、免疫组化等方法,观察地塞米松、齐留通对模型鼠5-LOX基因、蛋白表达的影响。结果哮喘组、地塞米松组肺组织中5-LOX mRNA、蛋白表达显著高于正常组、齐留通组(P<0.05);齐留通+地塞米松组5-LOX mRNA表达显著低于齐留通组(P<0.05)。结论糖皮质激素不能抑制5-LOX基因表达、蛋白合成;齐留通可抑制5-LOX基因表达、蛋白合成;糖皮质激素、齐留通联用较齐留通单用具有更强的抑制5-LOX作用。 Objective To investigate the effect of dexamethasone, zileuton on 5-1ipoxygenase (5-LOX) expression in the lungs in murine model of asthma. Methods The asthmatic model was induced by being sensitized and challenged with ovalbumin. By morphology examination, immunohistochemistry and in situ hybridization, we observed the regulation of dex- amethasone, zileuton on 5-LOX mRNA and protein expression. Results 5-LOX rnRNA and protein expression in asthma, dexamethasone group increased than control and zileuton group. Expression of 5-LOX mRNA in zileuton group was higher than dexamethasone plus zileuton group. Conclusion Glucocorticoids could not inhibit 5-LOX gene and protein expres- sion. 5-LOX gene and protein expression were inhibited by zileuton. Compared with zileuton alone, glucoeorticoids plus zileuton results in stronger effect on 5-LOX inhibition.
