摘要
目的:采用高效液相色谱法测定不同产地及不同采收期山茱萸中的莫诺苷、马钱苷及獐牙菜苷3个环烯醚萜苷类的含量。方法:采用Welchrom-C18(250 mm×4.6 mm,5μm)色谱柱,柱温35℃,以甲醇-0.1%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL.min-1,检测波长240 nm。结果:莫诺苷、马钱苷和獐牙菜苷的进样量分别在12.0~600.0,8.64~432.0,3.72~186.0μg.mL-1范围内,与色谱峰面积呈良好线性关系(r=0.9997,n=6);平均回收率(n=6)分别为96.3%,103.4%,98.8%。不同产地的山茱萸中,莫诺苷以西北大学果园采收的含量最高,马钱苷以陕西省丹凤县产的含量最高,獐牙菜苷以西北大学果园采收的含量最高;不同采集时间的西北大学果园山茱萸中莫诺苷、马钱苷和獐牙菜苷的含量范围分别为15.21~50.50,2.72~8.21,0.30~1.43 mg.g-1。结论:本方法操作简单、快速,重复性好,可同时测定山茱萸中莫诺苷、马钱苷和獐牙菜苷的含量。
Objective:To establish an HPLC method for simultaneous determination of three secoiridoid glucosides (morroniside, loganin and sweroside) in Fructus Corni from different sources and during different harvest periods. Methods: Welchrom C18 (250 mm × 4.6 mm, 5 μm) column was used and methanol and water( containing 1% phosphoric acid) solution as mobile phase in a gradient elution, the flow rate was 1.0 mL · min^-1, the detection wavelength was at 240 nm and the column temperature at 35 ℃. Results: The linear ranges of morroniside, loganin and sweroside were 12.0 -600. 0, 8.64 -432. 0, 3.72 - 186.0 μg . mL^-1μg, respectively(r =0. 9997,n =6). The average recoveries (n = 6 ) were 96.3 % , 103.4% , 98. 8 % , respectively. The content of morroniside of sample collected from botanical garden of Northwest University is the highest; The content of loganin of sample collected from Shaanxi Danfeng is the highest; The content of sweroside of sample collected from botanical garden of North- west University is the highest. The content ranges of morroniside, loganin and sweroside in Fructus Corni from dif- ferent harvest periods were 15.21 - 50.50, 2.72 - 8.21, 0. 30 - 1.43 mg .g^-1. Conclusions: The validated HPLC method has the advantages of precision, reproducibility and reliability, allowing simultaneous determination and comparison of morroniside, loganin and sweroside in Fructus Corni.
