摘要
【目的】探讨补肾祛风湿中药昆仙胶囊治疗类风湿关节炎(CIA)的作用机理。【方法】选用SPF级Wistar大鼠随机分为正常组,模型组,甲氨蝶呤(MTX)对照组(剂量为0.625 mg.kg-1.周-1)和昆仙胶囊(中药)低、中、高剂量组(剂量分别为0.2、0.4、0.8 g.kg-1.d-1)。采用尾根部皮下多点注射胶原蛋白法复制CIA大鼠模型,测量各组大鼠后肢关节肿胀度,采用酶联免疫吸附(ELISA)法检测大鼠血清中白细胞介素-8(IL-8)的含量,Real-time PCR法检测大鼠滑膜组织IL-8 mRNA的表达水平。【结果】与模型组比较,MTX组、中药各剂量组大鼠滑膜组织IL-8 mRNA表达均显著降低(P<0.05),并可显著降低滑膜组织病理积分(P<0.05或P<0.01);中药高剂量组大鼠血清中IL-8水平显著降低(P<0.05),并可显著降低软骨组织病理积分(P<0.01)。【结论】补肾祛风湿中药昆仙胶囊可能通过抑制大鼠滑膜及血清中IL-8的基因表达,达到抑制大鼠滑膜炎、控制类风湿关节炎炎症活动的目的,且治疗效果与剂量呈正相关。
Objective To explore the therapeutic mechanism of Kunxian Capsules, which have the actions of tonifying kidney and dispelling wind-damp, for collagen-induced arthritis ( CIA ). Methods Specific-pathogen free Wistar rats were randomized into normal control group, model group, methotrexate ( MTX, 0. 625 mg·kg^-1 per week) group, and low-, moderate- and high-dosage Kunxian Capsules groups (0.2, 0.4, 0. 8 g·kg^-1·, respectively). Multi-point subcutaneous injection of collagen protein into the caudal root was used for the induction of CIA. The pedal swelling of rat posterior limb was measured, and serum interleukin 8 (IL-8) content was detected with enzyme-linked immunosorbent assay (ELISA). Real-time polymerase chain reaction (PCR) was used for the detection of IL-8 mRNA in the synovia and serum of rats. Results Compared with the model group, IL-8 mRNA expression was decreased (P 〈 0.05 ) and synovial pathological scores were reduced in MTX group and Kunxian Capsules groups ( P 〈 0.05 or P 〈 0.01 ), serum IL-8 level was decreased obviously ( P 〈 0.05 ) and synovial pathological scores were reduced in high-dosage Kunxian Capsules group (P 〈 0.01 ). Conclusion The therapeutic mechanism of Kunxian Capsules for relieving rheumatoid arthritis is probably related with the inhibition
