摘要
以8个马铃薯品种(系)为试材,进行叶片离体再生研究。结果表明,云薯501愈伤组织诱导的最佳培养基为MS+6-BA 3mg/L+NAA 0.3 mg/L+GA310 mg/L,愈伤诱导率为100%,诱导不定芽分化的优化培养基为MS+6-BA 3 mg/L+GA310 mg/L,芽的分化率为86.3%;会-2的愈伤组织诱导的最佳培养基为MS+6-BA 3 mg/L+NAA 0.5 mg/L+GA310 mg/L,愈伤诱导率为95.7%,诱导不定芽分化的优化培养基为MS+6-BA 3 mg/L+NAA 0.02 mg/L+GA310 mg/L,芽的分化率为65.4%。上述2个品种适宜用作马铃薯转基因实验的载体,其他6个马铃薯品种(系)的愈伤诱导率很低,有的品种(系)没有愈伤组织产生,不能作为马铃薯基因转化的载体。
Tbe regeneration system of eight potato cultivars was studied with leafs as explants. The results showed that the optical callus in- duction medium was MS ±6-BA 3 mg/L ± NAA 0.3 mg/L ± GA3 10 mg/L for Yunshu 501 and MS ±6-BA 3 mg/L ± NAA 0.5 mg,/L ± GA3 10 mg/L for Hui-2, and the induction rate reached to 100 % and 95.7 %, respectively. The optical shoot induction medium was found to be MS ± 6-BA 3mg/L ± GA310 mg/L for Yunshu501 and MS ± 6-BA 3 mg/L ± NAA 0.02 mg/L ± GA3 10 mg/L for Hui-2, and the re- generation frequency reached to 86.3 % and 65.4 %, respectively.
