家蚕健康性分子检测研究

摘要 家蚕体内的热激蛋白数量与家蚕材料的健康性呈现有规律的变化,利用生物信息学分析方法筛选出具有代表性的家蚕热激蛋白基因(Bmhsp24.3)并对其进行实时定量(Real Time PCR)表达分析。结果表明,目的基因Bmhsp24.3在不同家蚕材料中都有所表达,但在不同材料间的表达差异很明显;饲育条件不同,该基因相对表达水平有着不同程度的变化,饲育温度的升高,该基因的表达量都有不同程度的提高;健康性好的材料,目的基因的表达量增加的较多,健康性差的材料,目的基因表达量增加的较少。目的基因Bmhsp24.3在不同材料间的表达差异,刚好与品种的健康性保持一致。本研究实验结果说明材料的健康性与目的基因Bmhsp24.3表达量存在相关性,目的基因Bmhsp24.3表达量越高,材料的健康性越好;反之,目的基因Bmhsp24.3表达量越低,材料的健康性越差。 The heat shock protein expressional quantity in the larvae ofBombyx mori presented regular change with the healthiness tithe silk- worms. The representative gene of Bmltsp24. 3 were screened out by hioinformatic analysis and the expressional change in fat body in different materials were analyzed by using real time PCR method. The result showed that the gene Brn^p24. 3 was expr~;ed in skin, fat body and silk gland of silkworm were found by using chip data,of which the expressional level were obviously different. Real-time PCR result revealed that the gene's expression in larval fat body d Bomby'x mori were remarkably different in different materials. The relative quantity was very dis- tinctly changed under different breeding condition. The expressional levels were differentially improved along with the rising temperature. The expression levels remarkably increased in good healthy materials and unconspicuous increased in worse healthy materials. The expres- sional level' s difference in different materials of Bomb~ mori were in accordance with their healthiness, and the results indicated that the healthiness of material was positive correlation with the gene's expressional level in different materials of Bombyx mori. The higher the target gene expressional level, the better the materials' healthiness was. On the contrary, the lower the target gene expressional level, the poorer the materials' healthiness was.
