眼镜蛇毒细胞毒素对S180荷瘤小鼠抗肿瘤作用的研究 被引量:2

摘要 目的:探讨眼镜蛇毒细胞毒素(Cytotoxin,CTX)对S180荷瘤小鼠的抗肿瘤作用。方法:采用昆明小鼠S180实体瘤模型,通过腹腔注射CTX,观察CTX对S180小鼠的肿瘤重量、亚显微结构、凋亡率和pro-caspase-3表达量的变化。结果:CTX对S180肿瘤细胞的生长表现出明显抑制作用。0.2、0.4、0.8mg/kgCTX剂量组的抑瘤率分别为23.8%、54.1%、63.2%;透射电子显微镜和流式细胞仪检测显示S180肿瘤细胞发生凋亡;免疫印迹结果显示pro-caspase-3蛋白表达降低,提示caspase-3被激活。结论:CTX通过诱导细胞凋亡来抑制S180肿瘤细胞的生长。 Objective:To investigate the antitumor activity of Naja naja atra venom cytotoxin in S180-bearing mice.Method:After the intraperitoneal injection of CTX in the S180-bearing Kunming mice,the changes of the tumor weight,ultrastructure,apoptosis rate and the expression of pro-caspase-3 were observed.Result:CTX showed significantly inhibition on the growth of S180 tumor cells.The inhibitory rates of CTX were 23.8 %,54.1 %,63.2 % at the dosages of 0.2,0.4,0.8 mg/kg,respectively.Transmission electron microscopy and flow cytometry showed that CTX induced S180 cell apoptosis.Immunoblotting showed that the expression of pro-caspase-3 protein was decreased,which suggest that caspase-3 protein is activated.Conclusion:CTX could inhibit the growth of S180 tumor cells by inducing cell apoptosis.
