生肌玉红胶原海绵促进胶原合成与组织愈合的研究 被引量:15

摘要 目的探讨生肌玉红胶原海绵促进肉芽生长与创面愈合的可能机理。方法修饰后胶原海绵、生肌玉红膏、生肌玉红胶原海绵敷于大鼠背部机械性皮肤全层创面,控制组以凡士林纱布覆盖,于造模后3、7、14d,测试创面愈合率、肉芽组织中Ⅰ、Ⅲ胶原含量、bFGF阳性细胞数、羟脯氨酸含量。结果造模第3天时,羟脯氨酸含量、bFGF阳性细胞数、Ⅰ型胶原含量,生肌玉红胶原海绵组均显著高于生肌玉红膏组及控制组;第7天时,实验组各指标均显著高于控制组,其中创面愈合率、Ⅰ型胶原含量、bFGF阳性细胞数、羟脯氨酸含量,生肌玉红胶原海绵组显著高于生肌玉红膏组;第14天时,Ⅰ型胶原含量各组无显著差异,创面愈合率、bFGF阳性细胞数、Ⅲ型胶原含量、羟脯氨酸含量,生肌玉红胶原海绵组显著高于控制组。结论生肌玉红胶原海绵促进创面愈合与减少瘢痕形成的机制之一可能是调节创面bFGF水平与调节创面Ⅰ、Ⅲ型胶原含量实现的。 OBJECTIVE To explore the possible mechanisms of promoting granulation tissue growth and wounds healing by Shengji Yuhong collagen sponge. METHODS Applying modified collagen sponge, Shengji Yuhong Ointment and Shengji Yuhong collagen sponge on the full layers of mechanical wounds on the dorsum of each SD rat respectively, and the control group were covered with Vaseline gauze at the same time. The wounds healing rate, the amounts of type I and III collagen contents in the granulation tissue, the number of positive bGFG cells and contents of hydroxyproline were evaluated on the 3rd, 7th, 14th day during modeling. RESULTS On the 3rd day of modeling, the content of hydroxyproline, positive bFGF cells and I collagen in Shengji Yuhong collagen sponge group were obviously higher than Shengji Yuhong Ointment group and control group. On the 7th day, the indexes of each experimental group were significantly higher than control group ; the wounds healing rate, type I collagen contefit, positive bFGF cells number and content of hydroxyproline in Shengji Yuhong collagen sponge group were especially much higher than Shengji Yuhong Ointment group. On the 14th day, there was no difference in type I collagen content among all the groups, while there were obvious differences between Shengji Yuhong collagen sponge group and Control group in wounds healing rate, positive bFGF cells number, type III collagen content and hydroxyproline content. CONCLUSION Shengji Yuhong collagen sponge has better therapeutic effects than traditional Shengji Yuhong Ointment in promoting wound granulation growth with less scar formation and conducive would healing by regulating the bGFG level, type I and Ⅲ collagen content.
