摘要 目的检测吴茱萸碱在293T细胞中是否激活AMP蛋白激酶(AMPK),并探索吴茱萸碱激活AMPK的可能机制。方法采用10%SDS-聚丙烯酰胺凝胶电泳及蛋白质免疫印迹检测磷酸化的AMPK和AMPK总量。将已知的AMPK激活剂(小檗碱、AICAR和A-769662)与吴茱萸碱同时处理293T细胞,通过检测吴茱萸碱与其共同处理的激活剂是否有协同效应来初步鉴定其激活AMPK的机制。结果本研究发现传统中药吴茱萸中的喹唑啉咔生物碱吴茱萸碱在293T中能激活AMPK,吴茱萸碱激活AMPK的机制不同于该研究中用到的AMPK激活剂(P<0.05,P<0.01)。结论吴茱萸碱是一种新AMPK激活剂,它通过不同于小檗碱、AICAR和A-769662的机制激活AMPK。 OBJECTIVE To investigate the mechanism by which evodiamine activates AMPK in 293T ceils. METHODS 293T cell were treated with evodiamine. 10% SDS-PAGE and Western Blot were used to examine AMPK phosphorylation and total AMPK. The mechanism of AMPK activation by evodiamine was investigated by examining the synergistic effects of evodiamine with other AMPK activators. RESULTS The results show that evodiamine activated AMPK in 293T cells. Evodiamine activated AMPK synergistically with berberine, AICAR or A-769992. CONCLUSION Evodiamine is a novel AMPK activator that activates AMPK by a mechanism different from that of AICAR, berberine or A-769992.
