摘要
本实验用 (1) 2 ,4 - D,(2 ) NAA/ BA和 (3) 2 ,4 - D/ KT3种生长调节剂组合对孝顺竹顶芽和节侧芽外植体进行愈伤组织的诱导。 2 ,4 - D诱导顶芽和节侧芽外植体几乎全部生出水质、粗糙的淡黄色愈伤组织 ;2 ,4 - D/ KT诱导节侧芽外植体半数生出颗粒状、光滑的白或淡黄色愈伤组织 ,不能诱导顶芽外植形成愈伤组织。由 2 ,4 - D诱导的孝顺竹愈伤组织在摇床上暗培养 ,建立了悬浮细胞系 ,可为竹子基因转化提供受体材料。对白绿竹和乌脚绿竹组织培养试管苗进行了移栽和生长观察 。
Calli were initiated in this experiment on the apical bud explants and nodal lateral bud explants in response to the combinations of three kinds of growth regulators,i.e.,2,4 D,NAA/BA,and 2,4 D/KT.2,4 D induced moist,rough,paleyellow calli on nearly apical bud and nodal lateral bud explants.2,4 D/KT induced smooth,granular,white calli on half nodal lateral bud explants and failed to induce calli on apical bud explants.When 2,4 D derived calli were cultured dark room on a gyratory shaker for more than 5 months,cell suspension line was developed,which can be used as the recipient material for bamboo gene transformation.The transplant and growth observation of tissue culture derived seedlings of Bambusa oldhamii and Bambusa edulis ,is helpful to the bamboo regeneration on tissue culture.
