摘要
以马铃薯幼嫩茎段为外植体,在相同激素条件下,于固体和液体培养基中附加不同浓度的氧化铕,探讨氧化铕的浓度及添加方式对马铃薯愈伤组织诱导及生长的影响。结果表明:不同浓度的氧化铕对马铃薯愈伤组织的诱导和生长的影响不同。统计学分析结果表明:固体培养基中氧化铕浓度为20.0~35.0 mg/L时,液体-固体双层培养时固体培养基中氧化铕浓度为10.0~30.0 mg/L时,液体-固体双层培养时液体培养基中氧化铕浓度为20.0~40.0 mg/L时,3种情况均适合马铃薯愈伤组织的诱导及生长。氧化铕在液体培养基中存在还是在固体培养基中存在对马铃薯愈伤组织的诱导和生长无明显影响,表明氧化铕的不同添加方式不会影响马铃薯愈伤组织的生长。扫描电镜结果也显示,适当浓度的氧化铕刺激马铃薯后,其愈伤组织细胞分裂速度更快。
The effect of Eu203 and addition manner(Eu203 in liquid or solid medium)on Solarium tuberosum callus, induced callus with young stems of potato as explants were studied. The results showed that the induction rate and growth of callus were affected by the Eu203 concentration. The callus grew well under the below conditions: 20.0-35.0 mg/L in solid medium, 10.0-30.0 mg/L in solid medium in liquid-solid medium nursing, and 20.0~ 40.0 mg/L in liquid medium in liquid-solid medium nursing. The induction and growth of callus were identical either Eu203 was added in liquid or solid medium.
