多巴胺受体D2型基因启动子区CpG岛甲基化状态与Ⅰ型和Ⅱ型精神分裂症的关联 被引量:3

摘要 目的探讨多巴胺D2受体(dopamine receptor D2,DRD2)基因启动子区CpG岛甲基化状态与Ⅰ型和Ⅱ型精神分裂症的关联。方法运用甲基化特异性PCR和直接测序法对101例Ⅰ型精神分裂症患者、82例Ⅱ精神分裂症型患者和100例健康被试DRD2基因启动子区CpG岛甲基化状态进行检测,分析三组的DRD2基因启动子区CpG岛甲基化状态差异。结果Ⅰ型精神分裂症组、Ⅱ精神分裂症组和健康对照组的DRD2基因启动子区CpG岛甲基化阳性率分别为:61.4%(62/101)、63.4%(52/82)和58.0%(58/100),差异无统计学意义;甲基化率分别为:0.38±0.17、0.31±0.13和0.32±0.14,三组甲基化率主效应显著,P<0.01。Ⅰ型精神分裂症组甲基化率高于Ⅱ型精神分裂症患者(P<0.01)和健康被试组(P<0.01)。后两者差异无统计学意义(P=0.793>0.05)。结论 DRD2基因启动子区CpG岛内位点高甲基化可能是Ⅰ型精神分裂症的发病机制之一。
