摘要
目的:检测贲门腺癌(gastric cardia adenocarcinoma,GCA)组织中Wnt通路相关因子Sox17基因的甲基化状态及mRNA表达情况,探讨其与贲门腺癌发生的关系。方法:应用甲基化特异性PCR(methylation specific PCR,MSP)及RT-PCR的方法检测126例贲门腺癌及相应癌旁非肿瘤组织中Sox17基因的甲基化状态及mRNA表达情况。结果:在126例贲门腺癌组织中,Sox17基因的甲基率为86.5%(109/126),明显高于癌旁非肿瘤组织(P<0.001);癌组织中Sox17基因的甲基化率与贲门腺癌的TNM分期及淋巴结转移均无关;癌组织中该基因mRNA的阳性表达率为58.7%(74/126),明显低于癌旁非肿瘤组织(P<0.001)。其mRNA阳性表达率与该基因的甲基化状态呈明显负相关(P<0.01)。结论:Sox17基因在贲门腺癌中呈高甲基化状态。
Objective: To investigate the state of methylation of the Sox17 gene in gastric cardia adenocarcinoma GCA ). Methods: Methylation-specific polymerase chain reaction ( MSP ) and real-time polymerase chain reaction ( RT - PCR ) methods were performed to examine the CpG methylation of the Sox17 gene and its mRNA expression in 126 GCA samples and in the corresponding paraneoplastic tissues. Results: The frequency of Sox1 7 gene methylation in the GCA tissues was 86.5 % ( 109 / 126 ), which is significantly higher than that in the paraneoplastic tissues ( P 〈 0.01 ). The hypermethylation of this gene showed no correlation with the clinical stage and pathologic classification of cancer tissues ( P 〉 0.05 ). The positive rate of Sox17 mRNA expression in the GCA tissues was 58.7 % ( 74 / 126 ), which is lower than that in the paraneoplastic tissues ( P 〈 0.01 ). The mRNA expression correlated with the frequency of Sox17 methylation ( P 〈 0.01 ). Conclusion: Sox17 methylation is a frequent event in GCA. Therefore, this gene can be included in the list of DNA-hypermethylated Wnt antagonist genes in GCA.
