半枝莲对人肝癌细胞株HepG2细胞黏附的影响

Effect of Herba Scutellariae Barbatae on Adhesion of Human Liver Cancer Cell Line HepG2

目的观察半枝莲对人肝癌细胞株HepG2细胞黏附的影响,探讨其抑瘤机制。方法 40只Wistar大鼠随机分为空白组及半枝莲小、中、大剂量组,分别早晚灌胃给药,半枝莲小、中、大剂量组给药量分别为0.75、1.50、3.00 g/mL,空白组给予等量生理盐水,连续4 d,第5日取血,分离血清,分别作用于等量HepG2细胞,MTT法检测各组细胞黏附情况。结果半枝莲组与空白组相比,能明显抑制HepG2细胞的黏附(P<0.05),呈明显的量效关系。结论半枝莲可能通过抑制肿瘤细胞的黏附来发挥抗肿瘤的作用。

Objective To observe the effect of Herba Scutellariae Barbatae on the adhesion of HepG2 cell,and explore its mechanism of anti-tumor.Methods Forty Wistar rats were randomly divided into the blank group and small-,medium-,large-dose drug group,and were administrated by gavage every day and night respectively.The small,medium,large dose was 0.75,1.50,3.00 g/mL respectively,the blank group was given the same amount of normal saline for 4 days.The serum was separated on the fifth day,to deal with the equivalent of HepG2 cells respectively.MTT method was used to detect the cell adhesion.Results Compared with the blank group,the drug group can inhibit HepG2 cell adhesion（P〈0.05）,it showed a significant dose-effect relationship.Conclusion Herba Scutellariae Barbatae develops its antitumor effect through inhibiting adhesion of tumor cell probably.
