HPLC同时测定云香精中苦参碱和氧化苦参碱含量

Simultaneous Determination of Matrine and Oxymatrine in Yunxiangjing by HPLC

目的建立同时测定云香精中苦参碱和氧化苦参碱含量的方法。方法高效液相色谱法,色谱柱为Agilent ZORBAX-NH2(4.6 mm×250 mm,5μm),流动相为无水乙醇-乙腈-3%磷酸溶液(9∶82∶9),流速为1.0 mL/min,检测波长为210 nm。结果苦参碱和氧化苦参碱线性范围分别为0.08～0.60、0.30～2.25μg,平均加样回收率分别为100.09%、100.10%,RSD分别为1.50%、1.55%。结论本方法准确、可靠、重复性好,能同时测定云香精中苦参碱和氧化苦参碱的含量,可用于云香精的质量控制。

Objective To set up a method for determining the content of matrine and oxymatrine simultaneously in Yunxiangjing.Methods HPLC method was used with Agilent ZORBAX-NH2 column（4.6 mm×250 mm,5 μm）.The mobile phase was anhydrous ethanol-acetonitrile-3%phosphoric acid solution（9∶82∶9）,and the detecting wavelength was at 210 nm.Results The linearity of matrine and oxymatrine was in the range of 0.08-0.60 μg and 0.30-2.25 μg,respectively.The average recovery was 100.09% and 100.10%,respectively.RSD was 1.50% and 1.55%,respectively.Conclusion This method is accurate and reliable with good reproducibility,and can be used for the simultaneous determination of matrine and oxymatrine in Yunxiangjing to control the quality.
