附子理中丸对正常及脾虚型溃疡性结肠炎模型大鼠长期毒性研究 被引量:6

摘要 目的:观察附子理中丸重复给药对正常及脾虚型溃疡性结肠炎模型大鼠的潜在毒性作用及靶器官,并比较两者的毒性大小。方法:大鼠随机分为正常组和模型组,各组均分为附子理中丸低剂量(7.5g/kg)、高剂量(15g/kg)和空白对照组(等容量蒸馏水),连续给药30天。观察一般行为和体征、血液细胞学、血液生化学及肉眼和光镜病理学。结果:模型组大鼠呈现脾虚症状,结肠病理学观察具有溃疡特征,附子理中丸灌胃给药30天后,正常给药组与正常对照比较,低剂HGB降低、CK升高;高剂T-BIL升高,左右睾丸指数降低;给药组PLT及LYMPH升高;BUN随剂量的增加而降低。模型给药与模型对照比较,高剂RBC及PLT升高、BUN及T-CHO降低;给药组CK升高。各剂量组内模型与正常比较,对照PLT、NEUT、脾指数升高,BUN降低;低剂NEUT、左睾丸指数升高;高剂CK,心、脾、左睾丸、左右肾及右睾丸指数升高;模型各给药组LYMPH及MONO升高。光镜下附子理中丸高剂组可见肾间质充血水肿及脊髓空泡变。结论:附子理中丸长期反复给药对大鼠肝肾功能和脊髓有一定的影响,正常动物与脾虚型溃疡性结肠炎模型动物间的毒性差异不明显。 To study the potential target organs and mechanism of long-termed repeated administration of Fuzi Lizhong Pills for normal and ulcerative colitis with weak spleen rats, and compare the toxicity between them. Methods: Rats were randomly divided into normal group and model group,two groups were each divided into pills 7.5g/kg, pills 15g/kg and blank control ( equal volume of distilled water) , continu- ous administration for 30 days. To observe the general behavior and signs, blood cytology, blood biochemistry and pathology with visual ob- servation and light microscopy. Results: The model rats have characteristics of symptoms of spleen, and features of colonic ulcers observed Compared with normal controls, HGB reduced and CK increased of low-dose; High-dose~ T-BIL increased; left and fight testicular index de- creased; treatment groups'PLT and LYMPH increased; BUN increased with the dose reduced. Models and model comparisons of administra- tion, RBC and PLT increased, BUN, and T-CHO reduced with high-dose; CK of all pills groups increased. Models within each dose group compared with normal, controls'PLT, NEUT, spleen index increased, BUN reduction; low-dose'NEUT, left testicular index increased; high doses CK, heart, spleen, left testicle, left kidney and right testis index rose high; Every treatment groups'LYMPH and MONO increased. Form Light microscope, founding high- dose groups change in the kidney and spinal cord, and no significant changes in other organs. Con- clusions : Fuzi Lizhong Pills with long-term and repeatedly dosing, may cause certain toxicity to liver, kidney and spinal cord, there is no sig- nificant difference between normal and ulcerative colitis with weak-spleen rats.
