Screening the antitumor effective fraction of acetic ether extracts seperated from amorphophallus Konja

摘要 目的:蛇六谷乙酸乙酯萃取物采用硅胶柱层析后的所得组分进行体外抗肿瘤活性筛选。方法:使用硅胶吸附层析分离技术,对蛇六谷乙酸乙酯萃取物进行化学成分分离;应用MTT法检测各组分对SGC-7901、HepG-2及C6细胞增殖的影响。结果:蛇六谷乙酸乙酯萃取物经硅胶柱层析分离,得到A1~A6,B1~B4,C,D等12个组分;MTT法实验显示A1、A2组分对SGC-7901、HepG-2及C6细胞的增殖均具有较强的抑制作用,并呈一定的量效关系。其中A1和A2在20μg浓度时,对HepG-2、SGC-7901、C6细胞增殖的抑制率分别达66.8%和68.4%;67.5%和77.8%;73.3%和70.6%。结论:蛇六谷乙酸乙酯萃取物经硅胶柱层析分离得12个组分中以A1、A2组分的抑瘤作用较强,为蛇六谷最佳有效部位。 To screen the antitumor effective fraction of acetic ether extracts seperated from Amorphophallus Konja in vitro. Methods: The various fractions of Amorphophallus Konja acetic ether extracts separated by technology of silica gel adsorption chromatography, which cell effects on proliferation of human stomach cancer SGC-7901 cells, hepatoma HepG-2 cells and rat glioma C6 ceils were detected by MTT method. Results: Twelve fractions obtained from Amorphophallus Konja ligarine extracts, which signed fraction A1 -A6, B1 -B4, C, D. The inhibition effects on ceils proliferation were observed on fraction A1 and A2 group and it showed some dose-effect relationship. Conclusion: Fraction A1 ,A2 have obvious antitumor effect in 12 fractions of acetic ether extracts seperatod from Amorphophallus Konja in vitro.
