表皮生长因子调节水通道蛋白3表达对结肠癌细胞迁移能力的影响 被引量:2

摘要 目的:探讨EGF调节AQP3表达对结肠癌细胞迁移能力的影响。方法:Western blot检测EGF不同条件作用下结肠癌细胞株HCT116 AQP3的表达情况;划痕实验检测不同AQP3表达程度下结肠癌细胞迁移能力的变化。结果:Western blot检测显示EGF上调AQP3表达并且呈时间和剂量依赖,在不同剂量EGF刺激下和不同作用时间下,AQP3的表达强度具有统计学差异(P<0.05);划痕实验示不同AQP3表达各组结肠癌细胞株HCT116迁移能力具有统计学差异(P<0.05),AQP3高表达可以促进结肠癌细胞迁移能力增强并且这一作用可以被AQP3抑制剂CuSO4阻断。结论:AQP3在EGF结合EGFR后上调表达并增强结肠癌细胞的迁移能力。 Objective:To investigate whether AQP3 expression in the human colon carcinoma cell lines enhances cell migration. Methods: The AQP3 expression level of colon cancer cell line HCT116 under different EGF condi- tions was analyzed by Western blot. Scratch assay test colon cancer cell migration under different AQP3 expression levels. Results: Western blot analysis showed that EGF increased the expression of AQP3 and was dependent on time and does. Scratch test showed AQP3 expression in different groups migration of colon cancer cell line HCT116 were significant differences ( P 〈 0.05 ). High expression of AQP3 can promote migration of colon cancer cells and this effect was blocked by AQP3 inhibitors CuSQ. Conclusion: After binding with EGFR, EGF up - regulated the ex- pression of AQP3. Over expression of AQP3 increase migration of colon cancer cells.
