摘要
目的:观察糙叶败酱大孔吸附树脂提取物(PsBe)对Sarcoma 180(S180)荷瘤小鼠的免疫调节作用。方法:建立S180腹水瘤动物模型,以环磷酰胺0.01g/kg作为阳性对照,分别给予PsBe 0.5g/kg、1.0g/kg、2.0g/kg治疗,观察荷瘤小鼠胸腺和脾脏脏器指数变化,MTT法测T细胞、B细胞转化功能,及NK细胞、LAK细胞活性及B细胞抗体产生能力,流式细胞术检测脾脏T细胞亚群。结果:PsBe 0.5~2.0g/kg可提高S180荷瘤小鼠脾脏指数和胸腺指数,增强T、B脾淋巴细胞转化功能和NK细胞、LAK细胞杀伤能力,提高B细胞抗体产生能力,增加脾T细胞亚群CD4+细胞和CD4+/CD8+比值,其中1.0g/kg组和2.0g/kg组与模型组比较,有显著性差异。结论:PsBe具有增强S180荷瘤小鼠免疫功能的作用。
To investigate the immune-regulatory effect of Patrinia scabra Bunge extracts (PsBe) separated by macroporous adsorptive resins on Sarcoma 180 transplanted mice. Methods: Sarcoma 180 ascites tumor animal models was established, cyclophosphamide 0.01 g/kg was choosen a positive control, PsBe O. 5g/kg, 1.0g/kg, 2. Og/kg were given as treatment. The change of thymus and spleen were ob- served after treatment. The activation of B and T cells, as well as the killing activity of NK and LAK ceils were evaluated by MTT method. In addition, the antibody producing ability of B cells in vivo was detected. Counting of T cells subsets was evaluated by flow cytometry. Re- suits: The spleen and thymus indexes of S180 tumor-burdened mice were improved by PsBe 0.5 ~ 2. Og/kg, the transformation functions'bf spleen T and B lymphocytes were strengthened, the killing abilities of NK and LAK cells were also prompted. Moreover, the antibodies pro- ducing ability of the B ceils in vivo was strengthened, the amount ratio of CD4+ cells to CD8+ cells was increased. Of which significant differences appeared in PsBe 1.0 and 2.0 g/kg group when compared with model group ( P 〈 O. 05 or P 〈 0.01 ). Conclusion : PsBe can increase the immune function of the S180 tumor-burdened mice.
