摘要
目的:研究黄连吴茱萸不同配比在Caco-2单层细胞模型中的跨膜转运,比较小檗碱等四种主要化学成分表观穿透系数Papp的异同。方法:复制Caco-2细胞模型,采用HPLC测定转运后药液5种化学成分含量,并计算吸收系数,考察黄连吴茱萸不同配比时小檗碱等四种化学成分在细胞模型中的吸收特征。结果:随着配比中黄连比例的增加,无论是AP-BL还是BL-AP,4种化学成分的Papp均随时间的推移呈线性增加趋势,即黄连:吴茱萸为6:1时吸收率最大,差异有显著性(P<0.01)。结论:增加黄连的配伍比例,有利于黄连和吴茱萸主要化学成份的吸收,黄连吴茱萸为6:1时各主要化学成分的吸收最多。
Objective: To study the membrane transport of couple of Coptis and Evodia in Caco-2 cell monolayer model,and compare the similarities and differences of apparent permeability coefficien(Papp) among four major chemical compositions.Methods: To establish Caco-2 cell monolayer model,and to determine the concentration of four kinds of chemical composition by(High-performance liquid) HPLC,then calculate the Papp value.Results: As the ratio of Coptis raises in the couple,both P(AP-BL) from apical side(AP) to basolateral side(BL) or P(BL-AP) from BL to AP,the Papp value of four kinds of chemical composition are increased linearly with time trend.Conclusion: To increase the proportion of Coptis,the main chemical ingredients of Coptis and Evodia will be absorbed better.Especially,the Papp is the largest when Coptis to Evodia is 6: 1.
