灯盏花素对D-半乳糖引起小鼠记忆障碍及肝、肾异常变化的影响 被引量:4

Effects of Breviscapine on memory deficits,liver and kidney abnormal changes induced by D-galactose in mice

摘要 目的:利用D-半乳糖引起小鼠衰老模型,观察灯盏花素(Breviscapine,Bre)对衰老小鼠记忆障碍、肝肾组织结构异常改变及血脂代谢的影响。方法:雌性昆明小鼠60只随机分为5个组,除空白对照组外,其他各组小鼠每天皮下注射D-半乳糖120mg/kg制备衰老模型,从第41d开始,3个剂量药物组小鼠在sc D-半乳糖的同时,腹腔注射Bre 5,10和20 mg/kg 67d,在给药第66d进行避暗箱实验训练,24h后进行小鼠记忆能力检测,在给药第67d避暗实验结束后小鼠断颈处死,取血清测定血脂变化,分离肝脏和肾脏HE染色观察其组织结构变化。结果:D-半乳糖明显引起小鼠记忆障碍、肝肾组织结构异常和血脂紊乱。与空白对照组相比,模型组小鼠避暗实验潜伏期缩短(P<0.01)、受电击次数多(P<0.01),血清中甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白(LDL-C)升高(P<0.01)、高密度脂蛋白(HDL-C)降低(P<0.05),肝、肾组织结构异常改变。5,10和20mg/kg灯盏花素给药67天可不同程度地降低D-半乳糖引起小鼠避暗实验潜伏期缩短和受电击次数的增多,改善血清血脂水平和肝、肾组织结构异常变化。结论:灯盏花素对D-半乳糖引起小鼠记忆障碍、血脂紊乱及肝肾组织结构异常改变具有明显的改善作用,该作用提示灯盏花素可能有利于延缓衰老和老年性痴呆病的治疗。 Objective: To study the improving effects of Bre on memory deficits,liver and kidney organ structure abnormal changes and blood lipid disorders induced by D-galactose in mice.Methods: KM mice were divided into five groups.The mice were daily given 120 mg/kg D-galactose for 67 days by subcutaneous injection(sc) but control group mice.On the day 41 of D-galactose given,the three drug group mice were daily administered Bre at doses of 5,10 and 20mg/kg by intraperitoneal injection(ip) and simultaneously given D-galactose with sc.On the day 66 of given D-galactose,the mice were trained by dark-box test and detected memory ability after 24h training.On the day 67 of given D-galactose,the mice were killed by decapitation after dark-box test.The serum was collected for determination of blood lipid and the liver and kidney were separated for morphological observation with HE staining.Results:D-galactose(120 mg/kg·d) significantly resulted in memory ability impairment,the blood lipid disorder and the liver and kidney morphological structure abnormal changes in mice.Compared with the control group mice,the latency of model group mice was significantly shorten(P0.01) and the electric shock number was obviously increased(P0.01) in the mice dark-box test,the serum TG、TC and LDL-C were marked increased(P0.01) and HDL-C was notable decreased(P0.05),and the liver and kidney morphological structure was marked changed.However,the three doses of 5,10 and 20mg/kg Bre can lower the short latency and increased electric shock number,improve the blood lipid disorders and of liver and kidney morphology abnormal changes.Conclusion:Bre can ameliorate the memory deficits,blood lipid disorders and the liver and kidney tissue abnormal changes induced by D-galactose in mice,which may be beneficial in delaying the aging and treating the senile dementia.
