糙叶败酱大孔吸附树脂提取物对小鼠Sarcoma 180移植瘤的作用 被引量:4

摘要 目的:观察糙叶败酱大孔吸附树脂提取物(PsBe)对Sarcoma 180(S180)荷瘤小鼠的抗肿瘤作用。方法:建立S180腹水瘤和实体瘤动物模型,分别给予PsBe 0.5g/kg、1.0g/kg、2.0g/kg治疗,观察荷瘤小鼠生命延长率和抑瘤率,HE染色观察肿瘤组织形态学变化,透射电镜观察肿瘤细胞结构变化,流式细胞仪检测Bcl-2、Bax、P53蛋白表达。结果:PsBe 0.5~2.0g/kg可提高S180腹水瘤荷瘤小鼠半数存活时间(P<0.01);PsBe 2.0g/kg显著抑制S180实体瘤生长,提高荷瘤小鼠生存质量(P<0.05);HE染色显示PsBe各组出现不同程度的肿瘤细胞减少;电镜下观察,PsBe各剂量组可引起肿瘤细胞凋亡;此外,PsBe1.0g/kg和2.0g/kg可增加P53蛋白表达,上调Bax/Bcl-2比例。结论:PsBe具有抑制肿瘤S180生长的作用,其作用与引起肿瘤细胞凋亡相关。 Objective: To investigate the anti-tumor effect of Patrinia scabra Bunge extracts(PsBe) separated by macroporous adsorptive resins on Sarcoma 180 transplanted mice.Methods: Sarcoma 180 ascites tumor and solid tumor animal models were established,PsBe 0.5g/kg,1.0g/kg,2.0g/kg were given as treatment.The life span and inhibition rate of tumor-bearing mouse were assessed after treatment.Tumor histological changes and cellular structure were observed by HE staining and Transmission Electron Microscopy(TEM) respectively.Bcl-2,Bax and P53 proteins were detected by flow cytometry.Results: Half survival time of mice bearing sarcoma 180 ascites tumor was significantly prolonged to varying degrees by PsBe from 0.5g/kg to 2g/kg(P 0.01).Not only the growth of Sarcoma 180 solid tumors was dramatically inhibited,the quality of life of tumor-bearing mice was also significantly improve by PsBe 2.0g/kg(P 0.05).HE staining showed tumor cells were decreased with different degrees in various PsBe groups.Apoptosis of tumor cells in PsBe groups can be observed under electron microscopy.In addition,P53 protein was increased and Bax/ Bcl-2 ratio was upregulated by PsBe 1.0g/kg and 2.0g/kg groups.Conclusion: PsBe can inhibit the growth of Sarcoma 180 tumor which related to it's cell apoptosis inducement effect.
