摘要
目的 :观察溃结灵对溃疡性结肠炎(UC)大鼠模型结肠黏膜核因子κB抑制蛋白-(I B)及I B激酶β(IKKβ)的蛋白表达作用,对其抗UC作用机制进行探讨。方法:采用三硝基苯磺酸(TNBS)法复制UC大鼠模型并进行溃结灵药物干预。采用Western blot方法检测结肠黏膜I B、IKKβ蛋白相对表达量。结果:模型组I B蛋白相对表达量明显低于正常组(P<0.05),溃结灵高剂量组(18.3g/kg)I B蛋白相对表达量显著高于模型组(P<0.01),中剂量组(9.2g/kg)I B蛋白相对表达量明显高于模型组(P<0.05);模型组IKKβ蛋白相对表达量显著高于正常组(P<0.01),溃结灵高(18.3g/kg)、中(9.2g/kg)两个剂量组IKKβ蛋白相对表达量均明显低于模型组(P<0.05)。结论 :溃结灵对TNBS法UC大鼠模型结肠黏膜I B蛋白表达的上调作用及对IKKβ蛋白表达的抑制作用,可能是其抑制NF-κB的活化,减轻炎症反应的作用机制之一。
Objective: To observe the effect of Kuijieling decoction(KD)on protein expression of I B and IKKβin colonic mucosa of rats with ulcerative colitis(UC)and to explore its possible mechanism.Methods :Trinitrobenzenesulfonic acid(TNBS)method was used for the establishment of UC rat model.After treatment with different dosages of KD,Western blot technique were used to detect protein expression of I B and IKKβin Colonic Mucosa.Results: The relative expression of I B protein in model group was obviously lower than that in normal group(P0.05),the relative expression of I B protein in high dosage group of KD was significantly higher than that in model group(P0.01),the relative expression of I B protein in middle dosage group of KD was obviously higher than that in model group(P0.01);The relative expression of IKKβprotein in model group was significantly higher than that in normal group(P0.01),the relative expression of IKKβprotein in both high and middle dosage groups of KD were significantly lower than that in model group(P0.05).Conclusion: The relative expression of I B protein is enhanced and the relative expression of IKKβprotein is Inhibited in colonic mucosa of UC model rats induced by TNBS after treatment with KD,Its possible mechanism may inhibit the activation of NF-κB and reduce inflammatory reaction.
