摘要
目的观察乙烯利对成年及子代雌性Balb/c小鼠体液免疫及细胞免疫的影响。方法成年动物以乙烯利灌胃染毒,剂量为15.9、47.7和143 mg/kg,4周后观察小鼠免疫器官脏器系数、外周血淋巴细胞及免疫功能的变化;成年雌性小鼠合笼后,于孕期第6天(GD6)开始灌胃染毒,至仔鼠断乳,仔鼠6周龄时观察仔鼠免疫器官脏器系数、外周血淋巴细胞及免疫功能的变化。采用LDH法测定自然杀伤(NK)细胞活性,采用T淋巴细胞增殖和迟发型变态反应来评价细胞免疫功能,采用溶血空斑实验来评价体液免疫功能。结果各剂量组成年小鼠及仔鼠体重及脏器系数未发生改变;143 mg/kg乙烯利能够使仔鼠淋巴细胞数增多,而成年小鼠未产生变化;143mg/kg的乙烯利能够抑制成年小鼠的NK细胞活性,但是47.7~143mg/kg的乙烯利却增强仔鼠的NK细胞活性;15.9~143 mg/kg乙烯利能够增强雌性小鼠T淋巴细胞增殖能力,但是子代小鼠各剂量组的T淋巴细胞增殖能力却未受影响;成年小鼠各剂量组迟发型变态反应均未发生显著变化,47.7~143 mg/kg浓度的乙烯利能够抑制仔鼠迟发型变态反应的发生;成年及仔鼠的体液免疫功能未受影响。结论乙烯利在15.9~143 mg/kg时能够影响成年及子代Balb/c小鼠的免疫系统,成年及子代小鼠免疫系统对乙烯利的反应性存在差异,表明同一化学物的免疫毒性及发育免疫毒性表现不完全相同;评价外源化学物对免疫系统的影响时,有必要时同时观察外源化学物对免疫系统及发育免疫系统的影响。
Objective To observe the effect of ethephon on cellular immunity and humoral immunity of adult and offspring female Balb/c mice.Methods Ethephon was gavaged to adult mice for 4 weeks at dosage of 15.9,47.7 and 143 mg/kg.After 4 weeks,mice were sacrificed to detect the immune function;Pregnant mice were gavaged by Ethephon at dosage of 15.9,47.7 and 143 mg/kg till the offspring weanling.The immune function was evaluated with NK cell activity,Tlymphocyte proliferation,PFC and DTH.At the same time,relative organ weight was recorded and anti-agglutination blood was collected to observe the condition of blood cell count.Results The lymphocyte percentage in offspring mice was increased by ethephon at the dose of 143 mg/kg,meanwhile there was no obvious effect in adult mice.The most striking difference between adult and offspring mice was the NK cell activity,Ethephon at dosage 143 mg/kg could inhibit NK cell activity of adult mice while 47.7~143 mg/kg Ethephon could enhance offspring’s NK cell activity.Ethephon could enhance T lymphocyte proliferation of adult mice.15.9~143 mg/kg ethephon did not influence the DTH of adult mice,however,47.7~143 mg/kg Ethephon inhibited DTH of offspring.The PFC result indicated that humoral immunity of adult and offspring mice were not affected.Conclusion Ethephon presented certain immunotoxicity and developmental immunotoxicity in dosage from 15.9 to 143 mg/kg.The character of immunotoxicity was different between adult and offspring mice.
