摘要
目的:研究不同培养条件对骨骼肌细胞增殖及分化的影响。方法:以体外培养小鼠肌母细胞C2C12为研究模型,对照组用含φ=10%胎牛血清液培养,实验组培养基改变为φ=2%胎牛血清的DMEM液,分别于更换培养液后24、48、72h,观察细胞形态变化。结果:对照组细胞形态实验前后无变化,实验组24、48h时部分细胞出现死亡,72h存活细胞开始融合,肌管形成。结论:胎牛血清含量为φ=2%的DMEM液有促进肌母细胞分化,从而进一步形成肌管的作用;含φ=10%的DMEM不能使之分化。
?Objective:To investigate the effects of different concerntration of fetal bovine serum(FBS) on the proliferation and differentiation of skeletal muscle cells. Methods: Murine C2C12 myoblasts were used in this experiment, the cells of control group were maintained in Dulbeccco's modified eagle's medium(DMEM)supplemented with 10% FBS,those of experimental groups were in DMEM with 2% FBS .Morphology of the cells was observed with phase contrast photomicroscopy 24, 48 and 72 hours after set up of the cultures. Results: No morphological changes wave observed in the control group; floating cells were observed in the 24 and 48 hour cultures, and multinucleated myotubes were observed in the 72 hour cultures in the experimental group. Conclusion: DMEM supplemented with 2% FBS can induce differentiation of myocytes and formation of multinucleated myotubes in vitro .
