摘要 目的:探讨磁导向甲氨蝶呤缓释药物(FM-MTX)的形貌特征及缓释特性。方法:采用分光光谱法、扫描电镜、透射电镜、X射线能谱分析等手段检测磁导向甲氨蝶呤缓释药物的含量及组成,并对FM-MTX的形貌特征及缓释特性进行检测。结果:化学交联法制备出磁导向甲氨蝶呤白蛋白缓释药物,其药物含量稳定。颗粒直径约(0.8±0.14)μm。在梯度磁场下比磁矩σs为0.9A·m2/kg。在0.9%氯化钠及RPMI1640培养液6h仅有30.5%药物释放到介质中,24h释药量约60%,48h接近78%。结论:以化学交联法制得的磁导向甲氨蝶呤缓释药物FM-MTX含量稳定。 Objective: To study the morphology and slow release characters of magnetic targeted ferromagnetic albumin methotrexate conjugates (FM MTX) as an antitumor drug.Methods:The morphology and slow release characters of FM MTX were studied with ultraviolet spectrophotometry,transmission electron microscope,scanning electron microscopy and energy dispersive analysis of X ray.Results:Particle diameter of FM MTX was (0.8±0.14) μm.The magnetic strength of FM MTX σ s was 0.9 A·m 2/kg. About 30.5 % of MTX were released into mediums by 6 h,about 60% by 24 h,approximately 78% by 48 h.Conclusion:The pharmaceutical parameters of FM MTX are stable with slow release characters within 48 h in vitro .
