摘要
目的 了解被LPS激活的裸鼠腹腔巨噬细胞在用双歧双歧杆菌的完整肽聚糖刺激后产生的NO、iNOS及cGMP的水平。方法 以Griess试剂、激光共聚焦显微镜以及放免法分别测定裸鼠腹腔巨噬细胞产生的NO、iNOS及cGMP的水平。结果 完整肽聚糖注射组裸鼠腹腔巨噬细胞在LPS诱导下产生的NO、iNOS及cGMP含量均显著高于对照组(P<001)。结论 双歧双歧杆菌的完整肽聚糖能协同LPS激活巨噬细胞,使之分泌多量的NO、iNOS及cGMP,这些信号效应分子介导了完整肽聚糖的多种重要生理功能。
Objective To study the levels of nitric oxide(NO), inducible nitric oxide synthase(iNOS) and cGMP produced by peritoneal macrophages of nude mice after stimulation by whole peptidoglycans of Bifidobacteria bifidum. Methods The content of NO, iNOS and cGMP derived from peritoneal macrophages of nude mice was detected by employing Griess reagent, laser scanning confocal microscopy and radioimmunoassay, respectively. Results The content of NO, iNOS and cGMP derived from peritoneal macrophages of nude mice induced by LPS in whole peptidoglycan injection group was significantly higher when compared with control group(P<0 01). Conclusions Whole peptidoglycans of Bifidobacteria bifidum in synergy with LPS could activate macrophages to produce a large amount of NO, iNOS and cGMP. These signal effectors mediated many kinds of significant physiological functions of whole peptidoglycans.
