摘要
目的: 用高效液相色谱法测定兔血清中琥乙红霉素的浓度。方法: 采用YWG- C18 色谱分析柱(4-6 m m×150 m m ,10 μm), 岛津SPD- 6AV 紫外检测器, 甲醇- 0-02 mol·L- 1 - 醋酸铵水溶液(70∶30 , 用氨水调pH7-3) 为流动相, 红霉素作内标。血液样品在碱性条件下用二氯甲烷提取纯化后进样, 在210 nm 波长处检测。结果: 琥乙红霉素线性范围为0-25~12 mg·L- 1 (r= 0-998 9), 最低检测量0-02 μg, 最低检出浓度0-05 mg·L-1 , 平均方法回收率96-26% 。精密度考察琥乙红霉素日内、日间相对标准偏差均小于6% 。结论: 本方法快速、准确、重复性好。
Objective:A HPLC method is described for the analysis of erythromycin ethylsuccinate in rabbit serum.Methods:The YWG-C 18 column(4 6 mm×150 mm,10 μm)was used.The mobile phase consisted of methanol -0 02 mol·L -1 ammonium acetate (70∶30,pH 7 3).Detection was at 210 nm.Results:Erythr omycin was used as achieved by internal standard.The ranges of calibration curves were 0 25~12 mg·L -1 ( r =0 998 9).Detection limits were 0 02 μg and the lowest detectable concentration of roxithromycin was 0 05 mg·L -1 ,while the average recovery of the method was 96 26%.The RSD of intra-day and inter-day assays all less than 6%.Conclusion:The method is quick,precise and good repeat.
