抑癌基因PTEN与FHIT在卵巢上皮癌中的表达及其相关性研究 被引量:4

A STUDY OF EXPRESSION AND SIGNIFICANCE OF PTEN AND FHIT IN EPITHELIAL OVARIAN TUMORS

摘要 目的通过检测PTEN与FHIT基因蛋白在卵巢上皮性癌、交界性肿瘤、良性囊肿、正常卵巢组织中的表达,探讨抑癌基因PTEN与FHIT基因蛋白在卵巢癌发生发展中的作用,分析PTEN与FHIT基因蛋白在卵巢癌中表达的相关性。方法采用免疫组织化学S-P方法检测卵巢上皮性癌45例、卵巢交界性肿瘤10例、卵巢良性囊肿10例、正常卵巢5例中PTEN、FHIT蛋白的表达。结果卵巢癌和卵巢交界性肿瘤组织中PTEN蛋白表达率(50.0%,53.3%)明显低于卵巢囊肿和正常卵巢组织(100%,100%);在卵巢上皮性癌中,PTEN蛋白表达与肿瘤的病理分级及临床分期呈负相关(P<0.05),与肿瘤的组织类型和淋巴转移无相关性(P>0.05);FHIT蛋白在卵巢上皮性癌中的表达明显低于交界性肿瘤、良性肿瘤及正常卵巢组织,差异有统计学意义(P<0.05);FHIT的阳性表达与肿瘤的组织分化正相关(P<0.005);而与临床分期、组织类型及淋巴结转移无相关性(P>0.05);PTEN和FHIT蛋白在上皮性卵巢癌中的共同联合表达有相关性(P<0.005)。结论 PTEN、FHIT基因蛋白在卵巢癌中表达显著降低,且二者表达呈正相关;患者卵巢肿瘤组织中的FHIT和PTEN基因蛋白表达显著降低,有望成为卵巢癌早期诊断的重要指标之一。 Objective To investigate the expression of PTEN and FHIT in epithelial ovariantissue in-cluding epithelial ovarian cancers, borderline tumors, benign tumors and normal ovary tissues. Further-more, to explore their relations, the clinicopathological characteristics and the potential role of PTEN and FHIT in epithelial ovarian cancer(EOC). Methods The expression of PTEN and FHIT proteins, using S -P immunohistochemical technique, were examined in 45 cases of epithelial ovarian cancers, 10 cases of borderline tumors, 10 cases of benign tumors and 5 cases of normal ovary tissues. The correlation between the expression of PTEN and FHIT proteins and clinicopathological characteristics in epithelial ovarian canc-er were analyzed. Results The positive rate of PTEN were 50% ,53.3%in borderline ovarian tumors and in EOC, they were significantly lower than that ovarian cyst and normal ovary. The expression of PTEN was negative relation to pathological type and pathological stages(P〈0. 005), no relation to tissue types and lymphatic metastasis(P〉0. 05). In different type ovarian cancer, the expression of PTEN was low trend, there was a significant different(P〈0.5). The positive expression of FHIT was relation to histologi-cal differentiation of ovarian eaneer(P〈0.005), but it was no relation to tissue types,lymph node metasta- sis and clinic pathological staging of EOC. The expression of PTEN and FHIT was in relationship. Conclu-sion The expression of PTEN and FHIT decrease in EOC, and the expression of them are positively cor- relation. The changes in the levels of that would be useful for early diagnosis of EOC.
