摘要
目的探讨款冬花挥发油对肺纤维化大鼠肺功能及细胞外基质代谢等的影响。方法采用博莱霉素气管注射建立大鼠肺纤维化模型。60只Wistar大鼠分为6组(每组10只):对照组、模型组、款冬花挥发油高、中、低剂量组及地塞米松组,以动物肺功能检测系统检测肺功能,以分光光度计检测肺组织中羟脯氨酸(Hyp)的含量。结果与对照组比较,模型组大鼠肺系数明显升高(P<0.01);吸气气道阻力(RL)、吸气总气道阻力(Re)及用力呼气时间(PET)显著升高(P<0.05或P<0.01);肺顺应性(Cdyn)、第0.3秒用力呼气容积(FEV0.3)、FEV0.3与用力肺活量(FVC)的比值(FEV0.3/FVC)、最大呼气中期流速(MMF)、用力最大呼气流速(PEF)显著降低(P<0.05或P<0.01);肺组织Hpy含量明显升高(P<0.01)。与模型组比较,款冬花挥发油高、中剂量组及地塞米松组的肺系数明显降低(P<0.05或P<0.01);款冬花高剂量组及地塞米松组RL、Re及PET显著降低(P<0.05或P<0.01);Cdyn、FEV0.3、FEV0.3/FVC、MMF、PEF增加(P<0.05或P<0.01);款冬花各剂量组及地塞米松组肺组织Hyp的含量明显降低(P均<0.01)。结论款冬花挥发油能够改善肺纤维化大鼠肺功能及肺组织细胞外基质的代谢。
Objective To investigate the effects of volatile oil of eohsfoot flower on lung function and metabolism of extracellular matrix in rats with pulmonary fibrosis. Methods Model of pulmonary fibrosis was established by injecting bleomycin in the airway. Sixty Wistar rats were divided into 6 groups (n = 10 each) : control group, model group, coltsfoot flower olatile oil high-, mid- and low-dose groups, dexamethasone group. Animal lung function was analyzed by animal lung function detection system, and content of hydroxyproline (Hyp) in lung tissue was assayed using spectrophotometer. Results Compared with control group, lung index, inspiratory airway resistance (RL) , inspiratory total airway resistance ( Re ), forced expiratory time (PET) and Hyp content of lung tissue were all significantly increased (P 〈 0. 05 or P 〈 0. 01 ) , and lung dynamic compliance (Cdyn) , forced expiratory volume in the first 0.3 second ( FEV 0. 3) , ratio of FEV O. 3 to forced vital capacity ( FEV 0. 3 / FVC) , maximal mid-expiratory flow rate (MMF) as well as peak expiratory flow rate (PEF) significantly decreased in model group( P 〈0.05 or P 〈0.01 ). Compared with model group, lung index in coltsfoot flower olatile oil high-, mid-dose groups and dexamethasone group significantly deereased(P 〈0.05 or P 〈0.01 ) ; RL, Re, PET sig- nificantly decreased( P 〈 0.05 or P 〈 0.01 ) ; FEV 0.3, FEV 0.3 / FVC, MMF as well as PEF were significantly increased in eoltsfoot flower olatile oil high-dose groups and dexamethasone group( P 〈0.05 or P 〈0.01 ). Hyp content of lung tissue significantly decreased in eoltsfoot flower olatile oil high-, mid-and low-dose groups and dexamethasone group ( all P 〈 0.01 ). Conclusions Volatile oil of cohsfoot flower could improve lung function and metabolism of extraeellular matrix in pulmonary fibrosis rats.
