全反式维甲酸和佛波酯对HL-60细胞表面N-糖链结构的影响

摘要 目的 :研究肿瘤细胞分化时细胞膜上糖链结构的改变。方法 :通过蛋白酶消化提取细胞表面3 H标记N 糖链 ,结合外切糖苷酶处理 ,经序列凝集素亲和层析分析HL 6 0细胞诱导分化后细胞表面N 糖链结构。结果 :全反式维甲酸 (ATRA)和佛波醇 12 豆蔻酰 13 乙酸酯 (PMA)均引起HL 6 0细胞二天线复杂型和高甘露糖型糖链升高 ,多天线复杂型糖链减少。但对平分型 β N 乙酰氨基葡萄糖 (GlcNAc)糖基的影响却相反 ,ATRA使之减低 ,而PMA则促进合成。结论 :在分化过程中 ,HL 6 0细胞表面N 糖链从高分支向低分支的转变 ,可能是某些肿瘤分化的一个特征。 Purpose:To study the structural changes of sugar chains on the cell surface during the differentiation of tumor cells.Methods: 3H labeled N linked sugar chains were extracted from the surface of HL 60 cells using pronase digestion after induced differentiation of the cells, and the structure of N sugar chains were analyzed with serial lectin chromatography,combining with exoglycosidase treatment. Results:After the treatment of all trans retinoic aicd(ATRA) or phorbol 12 myristate 13 acetate(PMA),the biantennary complex type and high mannose/hybrid type N sugar chains were increased,while the multi antennary sugur chain were decreased on the surface of HL 60 cells. However,the effects on bisecting GlcNAc residue were just opposite between ATRA and PMA. It was decreased by ATRA,but increased by PMA.Conclusions:During the course of cell differentiation,the structure of N sugar chains on the surface of HL 60 cells changes from high branching to low branching may be a characteristic of tumor cell differentiation. The bisecting GlcNAc is probably related to cell adhesion.
