胞嘧啶脱氨酶/氟胞嘧啶体系对人乳腺癌基因的实验治疗作用

摘要 目的 :研究胞嘧啶脱氨酶 (CD) /氟胞嘧啶 (5 FC)体系对人乳腺癌的实验治疗作用。方法 :应用MTT法测定人乳腺癌细胞对 5 FC的敏感性。结果 :5 FC对导入CD基因的人乳腺癌细胞有明显的细胞毒作用 ,而对未导入CD基因的人乳腺癌细胞的毒性较低 ,5 FC对导入和未导入CD基因的人乳腺癌细胞的IC50 分别为 418μg/ml和 142 9μg/ml。 结论 :CD/5 Purpose:To investigate the effect of cytosnie deaminase(CD)/fluorocytosine(5 FC) system on human breast Cancer.Methods:MTT assay was used to determine the sensitivity of 5 FC on human breast cancer cells.Results:It was found that significant cytotoxic activities were possessed by 5 FC on CD gene transduced breast carcinoma cells,but little effects on the non transduced breast carcinoma cells. The IC 50 values of 5 FC for CD gene transducecd and non transduced breast carcinoma cells were found to be 418?μg/ml and 1?249 ?μg/ml,respectively.Conclusions:The experiments indicate that CD/5 FC system has significant antitumor activities on the CD gene transduced breast carcinoma cells in vitro.
