摘要
目的 观察外源性视网膜母细胞瘤基因 (Rb基因 )对视网膜母细胞瘤 (retinoblastoma,RB)移植瘤细胞周期的影响。 方法 在建立裸鼠眼玻璃体腔 RB移植瘤模型及构建 Rb基因的逆转录病毒表达载体 PBabe- Rb的基础上 ,用脂质体 Dosper介导法将 Rb基因导入裸鼠 RB移植瘤 ,流式细胞仪检测 RB移植瘤细胞周期变化。 结果 Rb基因在 RB移植瘤内的表达使 RB移植瘤 G1 期细胞数明显增加 ,DNA指数及 S期百分比值有所降低。 结论 外源性 Rb基因可部分抑制 RB移植瘤细胞周期的进程。
Objective [WT5”BZ]To investigate the effect of exogenous Rb gene on the cell cycle of vitreous retinoblastoma (RB) transplantation tumor in nude mouse. [WT5”HZ]Methods [WT5”BZ]Based on establishing vitreous RB transplantation tumor in nude mouse,constructing retrovirus vector of Rb gene PBabe Rb and transfecing it into the RB transplantation model by liposome Dosper,the change of cell cycle of the RB transplantation tumor by flow cytometry(FCM)was analysed. [WT5”HZ]Results [WT5”BZ]FCM showed that the cells of G 1 phase of the treated eyes were obviously more than the control eyes with the value of DNA index(DI)and S phase fraction(SPF) decreased by the Rb gene expression. [WT5”HZ]Conclusion [WT5”BZ]The exogenous PBabe Rb gene can partially suppress the progress of the cell cycle of RB transplantation tumor in vivo. [WT5”HZ]
