摘要
目的 研究 NO在局灶性脑缺血再灌流过程中对神经细胞的作用机制 ,特别是 NO与细胞凋亡的关系。方法 利用流式细胞仪检测法 ,测定 N-硝基 -左旋 -精氨酸 (NNL A)干预后局灶性脑缺血鼠脑组织中神经细胞的凋亡情况。结果 在缺血再灌流期间过多或过少的 NO均对缺血区凋亡峰有明显影响 ,小剂量 NNL A减少神经细胞凋亡的发生率 ,大剂量 NNL A增加神经细胞凋亡的发生率。结论 局灶性脑缺血的不同区域 ,包括正常及病变组织中的凋亡细胞在数量及生物学意义上有很大的差别。不同剂量的
Objective To study mechanism of nitric oxide effecting on neuronal apoptosis (NO) in the focal cerebral ischemia and reperfusion,and relationship between NO and apoptosis. Methods Flow cytometry (FCM) was used to observe changes of neuronal apoptosis in the focal cerebral ischemia and reperfusion,experimental rats were treated with N nitro L arginine(NNLA) via intraperitineal injection. Results The increase or decrease of NO content effected the percentage of DNA fragmentation in the ischemic region distinctly. Small NNLA dose can be decreased incidence rate of neuronal apoptosis,high dose NNLA increased. Conclusion There is cell apoptosis in normal cerebral and ischemic penumbra and core of the infarction. The number and biology significance of apoptosis cell in these parts is distinction obviously. It is different that different dose of NO influenced on incidence rate of neuronal apoptosis.
