Oligodenderocyte and Oligodendroglioma Cells Secret Beta-trace Protein

摘要 目的:探素神经胶质细胞及人脑胶质瘤细胞是否能分泌β-tp。方法:用ELISA方法分析了胶质瘤病人脑脊液中和细胞培养液中β-tp水平。结果:3种胶质细胞瘤和患者脑脊液中均能测得β-tp,其浓度是正常人的10~12倍。培养的大鼠胶质细胞随细胞数增加而β-tp水平增高。在胶质瘤细胞株(SHG-44,U-251,BT-325)培液中β-tp浓度与细胞增殖的增加成正比。但在非胶质瘤细胞株,如卵巢癌细胞、神经母细胞瘤细胞及前列腺瘤细胞株等,β-tp的浓度与细胞增殖的趋势相反。结论:胶质细胞和人脑胶质瘤细胞能分泌β-tp,细胞分泌β-tp水平的高低由细胞类型及细胞的多寡决定。 Aim: To study levels of beta-trace protein (β-tp) in the cerebrospinal fluid with oligodendroglioma patients and in the mediums of incubated rat oligodenderocyte. Methods: Human brain cell of oligodendroglioma and non-oligoden- droglioma were analyzed by ELISA. Results: The level of β-tp in the cerebrospinal fluid with oligodendroglioma patients was 10-12 folds higher than that in the normal. Moreover, all of the incubated cells, including rat oligodenderocyte. human brain cell of oligodendroglionla and non-oligodendroglioma, could secret β-tp. However, there was a great difference in the level of β-tp secreted and in the relation of β-tp concentration and cellular proliferation among incubated carcinoma cells. The level of β-tp was in direct proportional to the proliferation of cell in the oligodendroglioma cell lines (SHG-44, U-251. BT- 325), but not in the non-oligodendroglioma cell lines (oophroma, prostate-carcinoma and neuro-oocyte). Conclusion: Oligo- denderocyte and oligodendroglioma cells in vitro were able to secret beta-trace protein and explained why the concentration of β-tp in CSF was higher than that in serum, it was suggested that the level of β-tp released depended on both of cell memher and type.
