摘要 目的 :建立血浆中呋塞米 (速尿 )简便敏感的HPLC测定方法。方法 :在低温下血浆经乙腈去蛋白后离心 ,采用反相C18柱及荧光检测 ,激发波长为 2 75nm ,发射波长为 410nm ,以 32 %乙腈 0 .0 5 %磷酸溶液为流动相 ,流速为 1.0ml·min-1,保留时间为 5min。结果 :呋塞米在 0 .0 2 5~ 10 μg·ml-1的范围内呈良好线性关系 (r =0 .9995 ) ,平均回收率为 (98~ 10 2 ) %。结论 :本法简便、灵敏、稳定 。 OBJECTIVE:To establish a simple,sensitive RP HPLC method of furosemide(FSM) determination.METHODS:The plasma was deproteinized with acetonitrile on ice,and then applied to HPLC using a reversed C 18 column and a fluorescence detector.The excitation wavelength of 275 nm and emission wavelength of 410 nm were used.The mobile phase was 32% acetonitrile 0.05% phosphoric acid at a flow rate of 1.0 ml·min -1 .The retention time was 5 minutes.RESULTS:A good linear relationship was found when FMS ranged from 25 ng·ml -1 to 10 μg·ml -1 , r = 0.999 5 ,average recovery varied from 98% to 102%.CONCLUSIONS:This method was simple,sensitive,stable and could use for clinical pharmacokinetic studies.
