摘要
目的建立和优化脑膜炎奈瑟菌(Neisseria Meningitidis,Nm)对人喉癌上皮细胞(Human Larynx Carcinoma Epithelial Cell,HEp-2)的黏附实验方法,检测C群序列型(SequenceType,ST)4821序列群和不可分群(Nongroupable,NG)Nm对HEp-2黏附能力的差异性。方法以C群Nm疫苗株C11作为靶菌,选用HEp-2,选择不同的黏附时间和不同的感染复数(Multiplicity of Infection,MOI)确定Nm与HEp-2黏附作用的最适条件,对中国分离的26株C群ST-4821序列群和8株NG Nm菌株进行HEp-2黏附能力研究。结果 Nm与HEp-2黏附研究最适作用时间为4h,最适MOI为100~1000;各血清群参考菌株的黏附菌落数在1.46×105~1.63×106菌落形成单位/毫升(Colony Form ingUnit,CFU/ml);26株ST-4821序列群的C群Nm对HEp-2的黏附菌落数在1.33×105~9.53×106CFU/ml;8株NGNm的黏附菌落数在3.04×105~8.52×106CFU/ml。结论不同血清群Nm对HEp-2的黏附能力存在差异;同是ST-4821序列群的C群Nm对HEp-2的黏附能力存在差异;同是NG Nm对HEp-2的黏附能力存在差异。
Objective To optimize the adhesive assay of Neisseria meningitidis, detect and analyze the adhesive ability of serogroup C and nongroupable in Neisseria meningitidis. Methods Neisseria meningitidis serogroup C strains, vaccine candidate strain (Ctl) was selected as targets. The adhesive colony forming unit (CFU) were determined in the different time and multiplicity of infection (MOI) using the larynx carcinoma epithelial cell line human larynx carcinoma epithelial cell (HEp-2).Then we determined the best time and MOI. The adhesive ability of 26 meningococcal group C sequence type(ST)-4821 complex isolates and 8 nongroupable isolates in Neisseria meningitidis were examined and compared by in vitro infection assays using the larynx carcinoma epithelial cell line HEp-2. Results The best adhesive time between Neisseria meningitidis and HEp-2 is 4h, while MOI between 100 and 1000 is suitable; adhesive ability of .different serogroups in Neisseria meningitidis were between 1.46 - 10^5 and 1.63 - 10^6 CFU/ml; adhesive ability of 26 meningococcal group C ST-4821 complex strains to HEp-2 were between 1.33 Y, 10s and 9.53 - 10^6 CFU/ml; adhesive ability of 8 serogroup nongroupable isolates in Neisseria meningitidis to HEp-2 were between.3.04- 10^5 and 8.52-10^6 CFU/ml. Conclusion Different serogroups in Neisseria meningitidis showed diversities of adhesive capacities. The adhesive ability of meningococcal groupC ST-4821 complex isolates is differential. Adhesive ability of serogroup nongroupable isolates in Neisseria meningitidis is different on the adhesive capacities.
