摘要
目的研究p,p’-DDE、β-六氯化苯(β-benzene hexachloride,β-BHC)联合染毒对青春期大鼠睾丸组织波形蛋白(Vimentin)、N-钙粘蛋白(N-cadherin)和卵泡刺激素受体(follicle stimulating hormone receptor,FSHR)mRNA和蛋白表达的影响。方法将20只50日龄清洁级雄性SD大鼠按体重随机分为4组,分别为对照(玉米油)组和p,p’-DDE(100 mg/kg)单独染毒组、β-BHC(100 mg/kg)单独染毒组、p,p’-DDE(100 mg/kg)+β-BHC(100 mg/kg)联合染毒组,每组5只。采用腹腔注射方式进行染毒,注射容积为10 ml/kg,隔天1次,连续进行10 d。采用Real-time PCR检测大鼠睾丸组织Vimentin、N-cadherin、FSHR mRNA的表达;采用Western Blot法检测Vimentin、N-cadherin、FSHR蛋白的表达。结果与对照组相比,β-BHC单独染毒组和p,p’-DDE+β-BHC联合染毒组大鼠睾丸组织Vimentin、FSHR mRNA的表达水平均下降,差异有统计学意义(P<0.05)。与p,p’-DDE+β-BHC联合染毒组相比,p,p’-DDE和β-BHC单独染毒组大鼠睾丸组织Vimentin mRNA的表达水平均较高,差异有统计学意义(P<0.05)。与对照组相比,p,p’-DDE和β-BHC单独和联合染毒组大鼠睾丸组织Vimentin、N-cadherin、FSHR蛋白的表达水平均下降,差异有统计学意义(P<0.05)。而p,p’-DDE和β-BHC单独和联合染毒组大鼠睾丸组织Vimentin、N-cadherin、FSHR mRNA和蛋白的表达水平间比较,差异均无统计学意义。结论 p,p’-DDE、β-BHC暴露可降低青春期大鼠睾丸组织中Vimentin、N-cadherin、FSHR的表达水平。
Objective To investigate the effects ofp ,p'-DDE and β-BHC on the expression of vimentin, N-cadherin and FSHR mRNA and protein in testes cells among pubescent rats. Methods Twenty male rats (50--day old) were randomly divided into four groups(five in each): p ,p '-DDE group(100 mg/kg), β-BHC group(100 mg/kg), p ,p'-DDE(100 mg/kg)+β-BHC (100 mg/kg) group as well as a control group treated with coin oil. The administrations were conducted by intraperitoneal injection, every other day for ten days, and the volume of injection was 10 ml/kg. The mRNA and protein expression levels of vimentin, N-cadherin and FSHR were measured by real time PCR and Western Blot, respectively. Results Compared with the control group, the mRNA levels of vimentin, FSHR in 13-BHC group and p ,p'-DDE(100 mg/kg)+β-BHC(100 mg/kg) group decreased significantly (P〈0.05); Compared with p ,p'-DDE (100 mg/kg)+β-BHC (100 mg/kg) group, the mRNA levels of vimentin increased significantly (P〈0.05) in the p ,p'-DDE group and β-BHC group. Compared witk the control group, the protein levels of vimentin, N-cadherin and FSHR decreased significantly (P〈0.05) in p ,p' -DDE grc,up, β-BHC group and p,p'-DDE (100 mg/kg)+β-BHC (100 mg/kg) group. Conclusion Exposure to p,p'-DDE and β-BHC can decrease the expression of vimentin, N-cadherin and FSHR in testes among pubescent rats.
