p16和HER-2蛋白在胃癌中的表达及其临床意义

Expressions and significance of p16 and HER-2 proteins in human gastric carcinoma

目的:探讨p16和HER-2蛋白在胃癌组织中的表达及其与临床病理参数的关系。方法:采用免疫组织化学S-P法检测60例胃癌及其30例胃正常组织中p16和HER-2蛋白的表达,并分析其与胃癌临床病理参数之间的关系。结果:p16在胃癌组织中的阳性率为21.7%,低于胃正常组织中的73.3%(P<0.01);p16在无淋巴结转移、Ⅰ～Ⅱ期胃癌患者的阳性率分别高于有高、中分化、Ⅲ～Ⅳ期患者(P<0.05和P=0.05)。HER-2蛋白在胃癌组织中阳性率为38.3%,高于在胃正常组织中的0.00%(P<0.01);HER-2蛋白在高、中分化、有远处转移和Ⅲ～Ⅳ期胃癌患者的阳性率分别高于低、未分化、无远处转移和Ⅰ～Ⅱ期患者(P<0.05～P<0.01)。p16和HER-2蛋白阳性之间无相关关系(P>0.05)。结论:p16蛋白的失表达和HER-2蛋白的过表达可能与胃癌的转移进展有关,对评估胃癌生物学行为及判断预后具有重要意义。

Objective：To investigate the expressions of p16 and HER-2 proteins in human gastric carcinoma tissues and its correlation with clinical pathological factors. Methods：The expressions of p16 and HER-2 proteins in 60 cases of gastric carcinoma and 30 cases of normal gastric tissues were detected using streptavidin-peroxidase immunohistoehemistry method, and the relation between the expressions and the clinicopathological parameter of gastric carcinoma was analyzed. Results：The positive rates of p16 in human gastric carcinoma and normal tissues were 21.7% and 73.3% , respectively （P 〈 0. O1 ）. The expression of p16 protein in gastric carcinoma tissues with no lymphatic metastasis and stage I - II TNM were higher than those with lymphatic metastasis （ P 〈 0.05 ） and stage m - 1v TNM（ P 〈 0.05 and P =-0.05 ）;The positive rates of HER-2 in human gastric carcinoma and normal tissues were 38.3% and 0. 00% ,respectively（P 〈 0.01 ）. The expression of HER-2 protein in gastric carcinoma tissues with high, middle differentiation, remote metastasis and stage ]]I- 1V TNM were respectively higher than those with no low, no differentiation, no remote metastasis or stage I - II TNM（P 〈0.05 -P 〈 0.01 ）. The expression of p16 protein was not correlated with HER-2 protein in human gastric carcinoma tissues（ P 〉 0.05 ）. Conclusions： Loss-expression of p16 and over-expression of HER-2 may be correlated with the metastasis and development of gastric carcinogenesis. Both of them are of great significance in evaluating the biological behavior and prognosis of gastric carcinoma.
