他克莫司结合蛋白5基因多态性与抑郁症的关联研究 被引量:1

Non-association between tacrolimus binding protein 5 gene polymorphisms and major depressive disorder in Chinese Han population

摘要 目的探讨他克莫司(FK506)结合蛋白5(FKBP5)基因多态性与抑郁症之间的关系。方法采用高温连接酶检测反应法,检测254例抑郁症患者(患者组)和231名正常对照者(对照组)的FKBP5基因rs3800373,rs13607802个位点基因型,分析基因型和等位基因频率在2组间的分布差异,及其与患者临床症状表型之间的关系。结果(1)患者组FKBP5基因rs3800373位点GG、GT、TT基因型频率分别为4.7%、41.9%、53.4%,对照组分别为6.6%、38.2%、55.3%,2组比较差异无统计学意义(X^2=1.252,P〉0.05);患者组FKBP5基因rs1360780位点CC、CT、TT基因型频率分别为55.4%、39.9%、4.7%,对照组分别为56.8%、37.0%、6.2%,2组比较差异无统计学意义(X。=0.739,P〉0.05);FKBP5基因rs3800373和rs13607802个位点等位基因频率在2组之间分布的差异无统计学意义(P均〉0.05);(2)单倍型分析显示,rs3800373-rs13607802个位点单倍型在患者组和对照组之间分布的差异无统计学意义(P均〉0.05);(3)rs3800373和rs13607802个位点不同基因型患者之间汉密尔顿抑郁量表、汉密尔顿焦虑量表和简明精神病评定量表各因子分值的差异均无统计学意义(P均〉0.05)。结论FKBP5基因rs3800373,rs1360780多态性可能与抑郁症缺乏关联。 Objective To investigate the association between tacrolimus binding protein 5 ( FKBP5 ) gene polymorphisms and major depressive disorder. Methods A total of 254 major depressive disorder patients and 231 healthy controls were recruited. Two polymorphisms (rs3800373 and rs1360780) of FKBP5 gene were genotyped using the ligase detection reaction. Results The distribution frequencies of the SNP of rs3800373 showed no significant differences between patients ( GG 4. 7% ; GT 41.9% ; TF 53.4% ) and controls (GG 6. 6% ;GT 38.2% ;TT 55.3% ) (X^2 = 1. 252,P 〉0. 05). The distribution frequencies of the SNP of rs1360780 showed no significant differences between patients ( CC 55.4% ; CT 39. 9% ; TY 4. 7% ) and controls(CC 56. 8% ;CT 37. 0% ;TT 6. 2% ) (X2 =0. 739,P 〉0. 05). No significant differences were found in allele frequencies of the SNPs of rs3800373 and rs1360780 between patients and controls ( P 〉 0. 05). No significant differences were found in rs3800373-rs1360780 haplotypes frequencies between patients and controls ( P 〉 0.05 ). Following analyzing the clinical symptom phenotypes, no association was found between the rs3800373 and rs1360780 polymorphisms and the factors scores of Hamihon depression scale (HAMD), Hamilton anxiety scale (HAMA) and brief psychiatric rating scale (BPRS) (P 〉0. 05 ). Conclusions The FKBP5 gene polymorphisms might not be associated with major depressive disorder.
