二陈汤对肺癌A549细胞中黏附分子-1和p38表达的影响

Effect of Erchen Decoction on Expression of ICAM-1 and p38 in Pulmonary Carcinoma Cell Line A549

目的研究二陈汤对肿瘤坏死因子-α(TNF-α)诱导的肺癌A549细胞中细胞间黏附分子-1(ICAM-1)和p38表达的影响。方法实验分为空白对照组、二陈汤对照组、TNF-α诱导组、二陈汤干预组、p38阻滞组,并给予相应干预。采用TNF-α诱导肺癌A549细胞产生ICAM-1高表达,以二陈汤含药血清处理ICAM-1高表达的肺癌A549细胞,采用Western blot免疫印迹法测定p38蛋白表达,以PCR法测定ICAM-1 mRNA水平,对二陈汤处理前后的ICAM-1表达水平及p38蛋白表达进行分析。结果二陈汤干预组肺癌A549细胞中血清ICAM-1表达及p38活性与TNF-α诱导组比较显著降低(P<0.01)。结论在体外实验中,二陈汤有可能通过抑制p38的活性而实现对ICAM-1表达的调控,这可能为二陈汤控制肺癌转移的机制之一。

Objective To study the effect of Erchen Decoction（ECD） on the expression of ICAM-1 and p38 protein in pulmonary carcinoma cell line A549.Methods A549 cells were divided into five groups：blank group,ECD control group,group induced by TNF-α,ECD treated group,p38 inhibitor-SB203580 group.A549 cells were treated by TNF-α to induce high expression of ICAM-1 and p38 protein,and then treated with ECD drug-containing serum and p38 inhibitor-SB203580.The expression of ICAM-1 was determined with RT-PCR,the expression of p38 protein was determined with western blotting.Results After treated by TNF-α,expression of ICAM-1 and p38 protein of A549 cells were highly induced compared with blank group（P〈0.01）.After treated by ECD and SB203580,expressions of ICAM-1 and p38 protein of A549 cells were obviously reduced（P〈0.01）,compared with group induced by TNF-α.Conclusion ECD can regulate the expression of ICAM-1 by inhibiting the activity of p38 protein in A549 cells,this may be one of the mechanism that ECD plays a role in anti-metastasis of lung.
