摘要
目的探讨三磷酸腺苷(ATP)对肌源性干细胞(MDSCs)凋亡的抑制作用及其机制。方法采用差速贴壁法分离新生小鼠MDSCs,利用40μg/L肿瘤坏死因子(TNF)-α诱导MDSCs凋亡,100txmol/LATP抑制细胞凋亡。经流式细胞仪检测MDSCs的凋亡率和半胱氨酰天冬氨酸特异性蛋白酶(Caspase)-3/8的表达。结果TNF-α诱导后,脱壁细胞明显增多,MDSCs的凋亡率由5.17%升至31.58%,增加了5倍,Caspase-3染色阳性细胞率由5.01%增至19.25%,Caspase-8染色阳性细胞率由2.97%增至8.58%。ATP作用后,MDSCs的凋亡率降至12.06%,减少了2/3,Caspase.3染色阳性细胞率降至9.86%,Caspase-8染色阳性细胞率降至6.96%,但仍高于对照组。结论ATP能抑制MDSCs表达Caspase-3/8,减轻TNF-α诱导的MDSCs凋亡。
Objective To explore the inhibitory effects of adenosine-triphosphate (ATP) on apop- tosis of muscle-derived stem cells ( MDSCs ) and the mechanism. Methods MDSCs were obtained from mouse skeletal muscle by preplate technique. The apoptosis of MDSCs was induced by 40 μg/L tumor necrosis factor (TNF)-α, and inhibited by 100 μmo]/L ATP. The apoptosis rate and the expression of Caspase-3/8 were measured by flow eytometer. Results As compared with that in group A, the apoptosis rate (31.58%) and the expression level of Caspase-3/8 proteins (19. 25%/8.58% ) were increased markedly in group C. As compared with that in group C, the apoptosis rate (12. 06% ) and ratio of Caspase-3 +/8+ MDSCs (9. 86%/6. 96% ) were decreased significantly in group D. Conclusion ATP could inhibit the expression of Caspase-3 and Caspase-8, which may reduce the TNF-α-induced apoptosis of MDSCs.
