抗抑郁药氟西汀对抗甲醛诱发炎性疼痛的作用

Effects of antidepressant fluoxetine on the formalin-induced inflammatory pain

摘要 目的观察抗抑郁药氟西汀的镇痛作用,并探讨其机制。方法小鼠左后掌心皮下注射6%甲醛溶液10μl制备炎性疼痛模型小鼠;造模前1h给予氟西汀灌胃,观察小鼠疼痛行为的变化,计算疼痛加权评分(PIS)。结果模型组一相PIS=(533.50±54.40)s,与模型组比较,氟西汀(35、70mg/kg)组PIS值明显下降[(198.28±62.71)s、(141.13±49.90)s];阿片受体阻断剂纳洛酮对此镇痛作用有一定的阻断作用,氟西汀与吗啡联合应用使镇痛效果加强。结论抗抑郁药物氟西汀对甲醛诱发的炎性疼痛有抑制作用;氟西汀与吗啡联合应用具有协同镇痛效应。 Objective To observe the analgesia and its mechanism of antidepressant iluoxetine. Methods Mice were intraplantarly injected with formalin (6%, 10 μl) in the right hindpaw after intra- gastric administration of fluoxetine. The time spent by the animals licking or biting the injected paw during 0-15 rain (first phase) and 15-60 min (second phase) was recorded and pain intensity scoring (PIS) was calculated. Results Fluoxetine dose-dependently inhibited the first fase response. PIS value in model group at the fisrt phase was (533.50 +54. g0) s, and fluoxetine (35 and 70 mg/kg) dose-dependently re- duced PIS value greatly [ ( 198.28 ± 62. 71 ) s and ( 141.13 ± 49.90) s]. The antinociceptive effect of fluoxetine was significantly attenuated in both phases by opioid receptor antagonist naloxone. Fluoxetine caused an augmentation in the analgesic effect of morphine. Conclusion The results indicate fluoxetine could antagonize formalin-induced pain and synergize analgesic effect with morphine.
