野生榆叶梅遗传多样性的AFLP分析 被引量:3

摘要 本研究利用AFLP分子标记技术,对野生榆叶梅6个居群进行分析。利用已筛选出的8对引物组合进行扩增,共产生256条谱带,其中206条谱带具有遗传多态性,占总谱带的80.47%;平均每对引物组合扩增产生129条多态条带,平均多态率为50.26%。6个居群观察等位基因数Na平均值为1.502 6,总Na值为1.8047;有效等位基因数Ne平均值为1.2165,总Ne值为1.2458;遗传多样性指数H平均值为0.1355,总H值为0.1567;Shannon多样性指数Ⅰ平均值为0.2134,总Ⅰ值为0.2554;野生榆叶梅6个居群有86.47%的遗传变异来自居群内,13.53%来自居群间,居群内遗传分化水平较高。AFLP分子标记聚类分析结果表明各居群基本按照地理距离进行聚类。 In this research, AFLP was employed to analyze genetic diversity of 6 wild populations of Prunus triloba. Eight pair primers previous identified were used to amplify specific DNA bands in the six populations. As a result, 256 DNA bands were generated and 206 of them were polymorphic bands, which was 80.47% of the total bands. Each primer pairs generated 129 polymorphic bands on average and the mean polymorphism rate was 50.26%. Among the six populations, the total observed number of alleles (Na) was 1.804 7 and the mean was1.502 6; the effective number of alleles (Ne) was 1.245 8 and the average was 1.216 5; the Nei's gene diversity index (H) was 0.156 7 and the average H was 0.135 5; the total Shannon's index (I) was 0.255 4 and the average was 0.213 4. The results of genetic differentiation indicates that 86.47% of the genetic variation should occure among populations while 13.53% should be within a population. The result of AFLP analysis reveals that dendrogram of 6 wild Prunus triloba populations is as similar as the clustering by geographic distance.
