摘要 目的研究Bcl-2在盐酸异丙肾上腺素(Isoprenaline hydrochloride,Iso)诱发大鼠心肌缺血早期的变化。方法注射盐酸异丙肾上腺素诱发大鼠心肌缺血模型。采用常规染色,免疫组化染色及RT-PCR方法分析Bcl-2基因和蛋白表达。结果 Bcl-2mRNA表达结果显示:注射Iso后12h,Bcl-2mRNA表达增加;注射24h后,Bcl-2mRNA表达与较12h有所下降,但相对于正常对照组(P<0.05)。Bcl-2蛋白表达随病变程度的加重而降低。结论心肌细胞凋亡是心肌缺血的基础,Bcl-2在早期(12-72h)便参与心肌缺血的全过程。 Objective To investigate the early change in the Iso-induced myocardial ischemia in rats.Methods Rat myocardial ischemia models were induced by isoproterenol(Iso)injection.Conventional staining,immunohistochemical staining and RT-PCR were used to analyze expression of Bcl-2 gene and protein.Results Bcl-2 mRNA arrived peak at 12 h(P0.05).Bcl-2 protein expression with the severity of disease decreases in 12-72 h.Conclusion Myocardial apoptosis is an important reason of Iso-induced myocardial ischemia,Bcl-2 had a hand in the whole process in the early myocardial ischemia.
