摘要
目的探讨文拉法辛对老年抑郁大鼠海马血管生成素-1(ANG-1)的影响。方法老年(20个月)雄性SD大鼠共36只,随机分为模型组、文拉法辛组和对照组,每组各12只。给予慢性不可预知的温和的应激结合孤养共5周,文拉法辛组于应激第3周末开始同时给予文拉法辛治疗持续14d,模型组同时给予生理盐水,对照组不给予任何处理。采用敞箱实验和蔗糖消耗实验评估大鼠抑郁模型。用免疫印迹(western blotting,WB)检测海马ANG-1蛋白表达,用逆转录聚合酶链反应(RT-PCR)检测ANG-1mRNA表达。结果①敞箱实验、蔗糖消耗实验显示模型组、文拉法辛组抑郁复制成功。②海马ANG-1表达情况:WB和RT-PCR显示,与对照组比较,文拉法辛组与模型组海马ANG-1蛋白及ANG-1mRNA均增加,差异有统计学意义(P<0.05);比较模型组,文拉法辛组ANG-1蛋白及ANG-1mRNA明显增加,差异有统计学意义(P<0.01)。结论慢性应激抑郁老年大鼠可能存在血管源性损害,文拉法辛可提高老年抑郁大鼠ANG-1表达,并可能对血管新生起到干预作用。
【Objective】 To study the effect of antidepressant Vanlafaxine works on angiopoietin-1(ANG-1) in hippocampus in elderly rat chronic stress model of depression.【Methods】 36 SD male rats including the elder(20 M) were randomly and divided into 3 groups: Venlafaxine,the model,the blank matched control.We adopted separation and chronic unpredicted stress to establish depression in all rats except those in blank matched control group for 5 weeks,and the depressive behavior of all animals was evaluated by open-field and fluid consumption test.ANG-1 expression were detected by Western Blotting and RT-PCR after treatment respectively with Venlafaxine or normal sodium at third weekend for 14 d.【Results】 Venlafaxine group and the model group were established depression model successfully assessed by open-field and fluid consumption test.ANG-1 expression in hippocampus was increased both in Venlafaxine group and in the model group,compared with the blank control group(P 0.05).After Venlafaxine administration for 14 d,ANG-1 expression in hippocampus was increased in Venlafaxine group compared with the model group(P 0.01).【Conclusion】 Vascular damage consists in the elderly depression after chronic stress,antidepressant Venlafaxine treatment increases ANG-1 expression in hippocampus of elderly rat depression model,and promote hippocampal angiogenisis.
