心房颤动患者血浆microRNA的表达差异 被引量:7

摘要 目的在心房颤动患者血浆中筛选出表达差异的microRNA(miRNA),寻找疾病相关的血浆生物标记物。方法采集100例心房颤动患者及100例对照组的血样标本,提取血浆中的miRNA,用Solexa测序方法在病例组及对照组各30人组成的样品混合池(pool)中筛选差异的血浆miRNA,分析测序结果并采用实时荧光定量聚合酶链反应对差异性显著的miRNA进行验证。结果 Solexa测序结果显示部分miRNA在心房颤动患者血浆中表达异常,心房颤动患者hsa-miR-409-3p表达量下降,与对照组相比差异有统计学意义(Z=-4.115,P<0.001);心房颤动患者hsa-miR-432表达量亦下降,与对照组相比差异有统计学意义(Z=-4.526,P<0.001);而心房颤动患者hsa-miR-328表达量下降与对照组比较,差异无统计学意义(Z=-1.234,P=0.217)。结论心房颤动患者血浆中存在miRNA表达差异,hsa-miR-409-3P、hsa-miR-432在患者血浆中低表达。 Objectives To find out the disease-related biomarkers by detection of plasm microRNA (miRNA) expression between the patients with atrial fibrillation (AF) and healthy people. Methods Blood sample of 100 patients with AF as case group and 100 healthy people as control group were collected to extract miRNA in plasma. The mixed pool of 30 patients" plasma miRNA was compared to the pool of 30 cases from control group by Solexa sequencing to screen significant differences of miRNAs, and then fluorescent quantitation polymerase chain reaction was used to verified the significant differences of miRNAs. Results There was part of the miRNA abnormal expression in patients with AF. The hsa-miR-409-3p, hsa-miR-432 expression was at a low level in patients with AF, and there was statistically significant compared to control group (respectively Z=-4.115,-4.526, both P〈O.O01 ), while the hsa-miR-328 expression deceased at no statistically significant compared to the control group (Z=-l.234,P=0.217). Conclusions Plasma miRNA expression differences exist in patients with AF, of which hsa-miR-409-3P, hsa[miR-432 with low expression.
