摘要
目的探讨人斯钙素(hSTC-1)在甲状腺乳头状癌患者外周血中的表达水平及其临床意义。方法采用逆转录-聚合酶链反应(RT-PCR)检测53例甲状腺乳头状癌患者和20例甲状腺良性肿瘤患者外周血中hSTC-1的表达水平,并与临床病理特征进行分析。结果 hSTC-1 mRNA在甲状腺乳头状癌患者外周血中的阳性表达率为39.62%(21/53),在甲状腺良性肿瘤患者外周血中则无阳性表达。甲状腺乳头状癌有淋巴结转移患者的hSTC-1 mRNA阳性表达率为66.67%,显著高于无淋巴结转移者的21.88%(P<0.05)。hSTC-1 mRNA在甲状腺乳头状癌患者中的表达与性别、年龄、临床分期无关。半定量水平检测显示,hSTC-1 mRNA在甲状腺癌患者外周血中的表达量与淋巴结转移有关,亦与性别、年龄、临床分期无关。结论应用RT-PCR检测外周血hSTC-1 mRNA有助于甲状腺乳头状癌患者的诊断和淋巴结转移的判断。
Objective To detect the expression of human stanniocalcin-1(hSTC-1) mRNA in peripheral blood of patients with thyroid papillary carcinoma,and analyze its clinical significance.Methods RT-PCR was used to detect hSTC-1 mRNA in the peripheral blood from 53 patients with thyroid papillary carcinoma and 20 patients with thyoid benign tumors.Its relationship with clinicopathological parameters were analyzed.Results The positive rate of hSTC-1 mRNA in 53 patients with thyroid papillary carcinoma was 39.62%(21/53),but there was no expression of hSTC-1 mRNA in 20 patients with benign tumors.The positive rate of hSTC-1 mRNA in patients with lymph node metastasis was 66.67%,while in patients without lymph node metastasis was 21.88%(P〈0.05).The hSTC-1 mRNA expression was not related to sex,age and clinical stages.The semi-quantitative RT-PCR demonstrated that the expression of hSTC-1 mRNA was related to lymph node metastasis,but had no relation with sex,age and clinical stage.Conclusion The detection of hSTC-1 mRNA in peripheral blood by RT-PCR has a high value for the determination of thyroid papillary carcinoma.The established method may serve as a useful objective parameter for identifying lymph node metastasis recurrence.
